基于超高效合相色譜對(duì)黃芪中5種主要黃酮類化合物的快速檢測(cè)

【摘要】:建立了利用超高效合相色譜法(Ultra performance convergence chromatography,UPC~2)快速分離和測(cè)定黃芪中5種主要黃酮類化合物(毛蕊異黃酮、毛蕊異黃酮苷、美的紫檀素、芒柄花苷、芒柄花素)的方法。黃芪樣品采用80%乙醇提取后,以超臨界CO_2-0.2%H_3PO_4-甲醇溶液為流動(dòng)相,梯度洗脫,色譜柱為Waters ACQUITY UPC~2CSH柱(100 mm×3.0 mm,1.8μm),柱溫為40℃,流速為0.4 m L/min,進(jìn)樣量為1μL,檢測(cè)波長(zhǎng)為280 nm。整個(gè)分析過(guò)程僅需15 min,分析速度是傳統(tǒng)液相色譜的3倍以上。結(jié)果表明:毛蕊異黃酮、毛蕊異黃酮苷、美的紫檀素、芒柄花苷與芒柄花素的檢出限及定量限范圍分別在0.3~0.5 mg/kg和1.0~2.0 mg/kg之間,5種黃酮類成分的加標(biāo)平均回收率均高于99.7%,相對(duì)標(biāo)準(zhǔn)偏差(RSD)小于2.2%(n=6);在最優(yōu)條件下,對(duì)13批不同產(chǎn)地的黃芪進(jìn)行檢測(cè),毛蕊異黃酮、毛蕊異黃酮苷、美的紫檀素、芒柄花苷與芒柄花素的含量范圍分別在4.8~102 mg/kg、14~277 mg/kg、0~135 mg/kg、5.3~119 mg/kg及2.8~41 mg/kg之間;本方法簡(jiǎn)便快速,重復(fù)性良好,結(jié)果準(zhǔn)確可靠,可用于黃芪藥材中5種主要黃酮類成分的含量測(cè)定。
[Abstract]:Ultrahigh performance performance convergence chromatography was established. Rapid separation and determination of five main flavonoids (isoflavone, isoflavone glycoside, rosewood and anthocyanin) from Astragalus membranaceus. Astragalus membranaceus samples were extracted with 80% ethanol, then gradient eluted with supercritical CO _ 2-0.2 and H _ 3PO _ 4-methanol solution as mobile phase.
