雙酚A對豚鼠的免疫毒性

【摘要】:為了研究BPA(bisphenol A,雙酚A)對豚鼠的免疫毒性,經(jīng)口灌服,以不同劑量(500、1 000 mg/kg)染毒14 d后接種O型口蹄疫疫苗,通過分析臟器系數(shù)、淋巴細(xì)胞增殖能力、NK細(xì)胞(natural killer cell,自然殺傷細(xì)胞)活性、特異性抗體水平以及細(xì)胞免疫水平,評價BPA染毒劑量對豚鼠體液免疫和細(xì)胞免疫的影響.結(jié)果表明:與陰性對照組相比,BPA染毒后豚鼠體質(zhì)量增加,心臟、肝臟、腎臟、腦、脾臟和卵巢體質(zhì)量也增加,相應(yīng)臟器系數(shù)升高;而胸腺、睪丸和附睪萎縮,相應(yīng)臟器系數(shù)降低.NK細(xì)胞數(shù)量雖有增加,但變化不顯著;BPA染毒引起NK細(xì)胞對靶細(xì)胞的殺傷能力顯著下降,并且隨著BPA染毒劑量的增加,其對靶細(xì)胞的殺傷能力顯著下降;淋巴細(xì)胞刺激指數(shù)顯著升高,并與染毒劑量呈正相關(guān).BPA對豚鼠的特異性抗體水平有一定抑制作用,并且隨著染毒劑量的增加該抑制作用愈發(fā)顯著.IL-2(白介素2,interleukin-2)和IFN-γ(干擾素-γ,interferon)水平低于陰性對照組,而IL-4(白介素4,interleukin-4)水平高于陰性對照組;隨著染毒劑量的增加,淋巴細(xì)胞增殖能力、IL-2和IFN-γ水平顯著降低,而IL-4水平顯著升高.研究顯示,BPA會損傷豚鼠的免疫器官,抑制機(jī)體產(chǎn)生體液免疫和細(xì)胞免疫,導(dǎo)致免疫功能紊亂.
[Abstract]:In order to study the immune toxicity of BPA(bisphenol A, bisphenol A (BPA) in guinea pigs, oral administration of different doses of 500 000 mg / kg for 14 days was used to inoculate O type foot-and-mouth disease vaccine, and the organ coefficients were analyzed. The activity of NK cell natural killer cell (natural killer cell), the level of specific antibody and the level of cellular immunity were evaluated. The effects of BPA dose on humoral and cellular immunity of guinea pigs were evaluated. The results showed that compared with the negative control group, the body mass of the heart, liver, kidney, brain, spleen and ovary also increased, and the organ coefficients of the thymus, testis and epididymis were atrophied. The number of NK cells increased, but the cytotoxicity of NK cells to target cells decreased significantly with the increase of dose of BPA, and the cytotoxicity of NK cells to target cells decreased with the increase of dose of BPA. The lymphocyte stimulating index (LSI) was significantly increased, and there was a positive correlation with the dose of exposure. BPA inhibited the specific antibody level in guinea pigs to some extent. With the increase of dose, the inhibitory effect of IL-2and IFN- 緯 was significantly lower than that of negative control group, but the level of IL-4 was higher than that of negative control group, and with the increase of dose, the level of IL-4 was higher than that of negative control group, and the level of IL-4 was higher than that of negative control group. The levels of IL-2 and IFN- 緯 decreased significantly, while the level of IL-4 increased significantly. It has been shown that BPA can damage the immune organs of guinea pigs, inhibit the production of humoral and cellular immunity, and lead to the disorder of immune function.
