油水淹地石油降解菌群的篩選和鑒定及高效菌群的構(gòu)建

【摘要】:從油水淹地污染土壤中獲得26株石油降解菌,篩選出表面活性劑產(chǎn)生菌1株(H-6)和優(yōu)勢(shì)菌6株(H-1、H-17、H-18、H-19、H-20、H-23),結(jié)合菌株形態(tài)觀察、革蘭氏染色和16S r DNA序列同源性分析,鑒定表面活性劑產(chǎn)生菌H-6為堀越氏芽孢桿菌(Bacillus horikoshii)。以表面活性劑產(chǎn)生菌H-6為中心,再任選3株優(yōu)勢(shì)菌株構(gòu)建石油降解菌群,得到高效石油降解菌群C5(H-1、H-6、H-18、H-19),通過正交試驗(yàn)得到菌群C5各菌種的最佳接種量。結(jié)果表明:接種量15%(H-1)、15%(H-6)、20%(H-18)、10%(H-19),溫度25℃,石油含量2 000 mg/L條件下,7 d時(shí)石油降解率達(dá)到78.87%。這說明高效降解菌群C5對(duì)石油具有較好的降解效果,可應(yīng)用于油水淹地污染土壤的修復(fù)。
[Abstract]:26 strains of petroleum degrading bacteria were obtained from contaminated soil under oil flooding, and 1 strain of surfactant producing strain H-6) and 6 strains of dominant bacteria were selected. Combined with morphological observation of strain, Gram staining and homology analysis of 16s r DNA sequence were carried out. The surfactant producing strain H-6 was identified as Bacillus horikoshiiformis. Taking the surfactant producing strain H-6 as the center, 3 dominant strains were selected to construct the petroleum degrading bacteria, and the high-efficiency petroleum degrading bacteria C5H-1H-6H-6H-18H-19 were obtained. The optimum inoculation amount of C5 strains was obtained by orthogonal test. The results showed that the oil degradation rate reached 78.87 at the temperature of 25 鈩,
