SRT對(duì)富集高聚PHA能力嗜鹽MMC的影響

【摘要】:調(diào)控菌群富集過(guò)程的工藝參數(shù),富集具有高PHA積累能力的混合菌(MMC)是發(fā)酵生產(chǎn)PHA的第一步也是最重要的一步。采用批式培養(yǎng)條件,集中研究了污泥停留時(shí)間(SRT)對(duì)PHA積累菌群的富集作用。研究結(jié)果表明SRT影響富集MMC的底物降解速率(qS),微生物積累PHA的速率(qP)和PHA積累能力。長(zhǎng)SRT(SRT=4 d)導(dǎo)致了較低的qS,qP和PHA細(xì)胞含量;過(guò)短的SRT(SRT=1 d)則降低了MMC的PHA轉(zhuǎn)化率(YPHA/S)和PHA積累能力,刺激了非PHA積累菌群的快速增殖。研究確定的最佳SRT為2 d,在此條件下富集的MMC最大PHA積累量可達(dá)細(xì)胞干重的56%。研究證明了SRT在選擇嗜鹽MMC中的重要作用,為快速富集PHA積累能力MMC奠定基礎(chǔ)。
[Abstract]:It is the first and most important step of fermentation to produce PHA by regulating the technological parameters of the enrichment process and enriching the mixed bacteria with high accumulation ability of PHA. The enrichment of PHA accumulation by sludge retention time (SRT) was studied by batch culture. The results showed that SRT affected the substrate degradation rate of MMC enrichment, the accumulation rate of PHA by microorganism and the accumulation ability of PHA. Long SRT(SRT=4 d) resulted in lower cell content of qSN QP and PHA, while too short SRT(SRT=1 d) decreased the PHA conversion rate of MMC, YPHA / S) and PHA accumulation ability, and stimulated the rapid proliferation of non PHA accumulative bacteria. The optimum SRT was determined to be 2 days. Under this condition, the maximum accumulation of MMC PHA could reach 56% of the dry weight of cells. The results show that SRT plays an important role in the selection of halophilic MMC and lays the foundation for rapid accumulation of PHA accumulation ability MMC.
