青霉素對(duì)土壤微生物群落結(jié)構(gòu)的影響

【摘要】:利用Biolog-ECO技術(shù)分析不同青霉素濃度處理下土壤微生物的群落代謝活性及結(jié)構(gòu)多樣性,利用變性梯度凝膠電泳(denaturing gradient gel electrophoresis,DGGE)技術(shù)分析典型微生物群落結(jié)構(gòu)變化情況,探討青霉素對(duì)土壤微生物群落的生態(tài)毒理效應(yīng)。主成分分析(PCA)結(jié)果表明,高濃度青霉素(400和800 mg·kg-1)處理能快速誘導(dǎo)土壤中以酚酸類化合物為碳源的微生物菌群富集。DGGE結(jié)果顯示,在高濃度青霉素(800 mg·kg-1)脅迫初期,土壤中微生物多樣性指數(shù)(Shannon-Wiener index)顯著降低(P0.05);7 d后,主要微生物群落結(jié)構(gòu)隨土壤中青霉素降解又逐漸恢復(fù)。
[Abstract]:Biolog-ECO technology was used to analyze the metabolic activity and structural diversity of soil microbes under different concentrations of penicillin. The changes of typical microbial community structure were analyzed by denaturing gradient gel electrophoresis (denaturing gradient gel electrophoresis, DGGE), and the ecotoxicological effect of penicillin on soil microbial community was discussed. The results of principal component analysis (PCA) showed that high concentration of penicillin (400 and 800 mg. Kg-1) could quickly induce the enrichment of microbiological bacteria in soil with phenolic acids as carbon sources, and the results showed that the microbiological diversity index (Shannon-Wiener index) in the soil decreased significantly (P0.05) at the early stage of high concentration of penicillin (800 mg. Kg-1). After 7 d, the main microbial community structure gradually recovered with the degradation of penicillin in soil.
