【摘要】：空氣污染是一種常見的現(xiàn)象.空氣污染物是誘發(fā)各類疾病的重要因素之一.以二氧化硫分子為代表,研究了它與Aβ17~42原纖維之間的相互作用.傘形抽樣模擬表明,二氧化硫分子的引入減少了纖維的解離自由能.分析各氨基酸殘基結(jié)合二氧化硫的頻數(shù)得知,二氧化硫分子更容易動(dòng)態(tài)地結(jié)合到疏水性氨基酸殘基的骨架上,同時(shí)也容易結(jié)合到帶電的氨基酸殘基上進(jìn)而削弱鹽橋相互作用,但是對這些相互作用的削弱是非常微小的,不足以破壞原纖維的結(jié)構(gòu).通過非線性主方程分析,我們發(fā)現(xiàn)二氧化硫分子加速了Aβ17~42二次成核過程,同時(shí)縮短了遲滯時(shí)間而加速纖維的聚集過程.通過這些計(jì)算,我們定量地揭示了空氣污染物與蛋白質(zhì)相互作用的特征,期望我們的結(jié)果為當(dāng)今社會(huì)評估污染物的健康效應(yīng)提供寶貴的參考信息.
[Abstract]:Air pollution is a common phenomenon.Air pollutants are one of the important factors inducing various diseases.The interaction between sulfur dioxide and A 尾 17N 42 fiber was studied.The Umbrella sampling simulation shows that the introduction of sulfur dioxide reduces the dissociation free energy of the fiber.By analyzing the frequency of the binding of amino acid residues to sulfur dioxide, it is found that sulfur dioxide molecules are more likely to bind dynamically to the framework of hydrophobic amino acid residues, and at the same time to the charged amino acid residues, thus weakening the salt bridge interaction.But the weakening of these interactions is too small to destroy the fabric of the primary fiber.Through nonlinear master equation analysis, we find that sulfur dioxide accelerates the process of A 尾 17N 42 secondary nucleation and accelerates the fiber aggregation process by shortening the hysteresis time.Through these calculations, we quantitatively reveal the characteristics of the interaction between air pollutants and proteins, and hope that our results will provide valuable reference information for the assessment of the health effects of pollutants in today's society.
