【摘要】：為揭示檸檬酸廢水生物處理過(guò)程中功能菌群作用機(jī)制,以檸檬酸工業(yè)廢水內(nèi)循環(huán)厭氧反應(yīng)塔(IC)中厭氧顆粒污泥為研究對(duì)象,統(tǒng)計(jì)顆粒粒徑分布,通過(guò)環(huán)境掃描電子顯微鏡(ESEM)觀察顆粒微觀形態(tài)結(jié)構(gòu),利用高通量測(cè)序技術(shù)分析微生物多樣性及菌群特征.結(jié)果發(fā)現(xiàn),粒徑在1.0~4.0 mm的顆粒所占比例最多,為74.4%.ESEM顯示微生物分布以球形細(xì)菌為主.高通量測(cè)序得到8 397條有效序列,可劃分操作分類單元(OTU)873個(gè),Alpha多樣性指數(shù)顯示樣品文庫(kù)覆蓋率0.936,Shannon指數(shù)為4.376,而ACE指數(shù)與Chao1指數(shù)分別為3 415.51與2 246.51,反映顆粒污泥中微生物種類與數(shù)量均較多.微生物菌群主要包括4大類,分別為可降解有機(jī)物的水解發(fā)酵菌群Paludibacter、Parabacteroides、Erysipelotrichaceae、Clostridium、Phascolarctobacterium、Aminobacterium、Saccharofermentans與Alkaliflexus(所占比例之和為24.93%);產(chǎn)氫產(chǎn)乙酸菌群Petrimonas與Syntrophomonas(所占比例之和為34.89%);產(chǎn)甲烷菌Methanosaeta(3.44%)及可耐受工業(yè)廢水毒害的微生物菌群Levilinea、Longilinea與Thermovirga(所占比例之和為14.62%).
[Abstract]:In order to reveal the mechanism of functional microflora in the biological treatment of citric acid wastewater, the anaerobic granular sludge in the inner circulating anaerobic reactor of citric acid industrial wastewater was used as the research object, and the particle size distribution was calculated. The microscopic morphology of particles was observed by environmental scanning electron microscope (ESEM), and the microbial diversity and microflora characteristics were analyzed by high-throughput sequencing technique. The results showed that the particle size of 1.0 ~ 4.0 mm was the largest. ESEM showed that the microbes were mainly globular bacteria, and 8397 effective sequences were obtained by high-throughput sequencing. The ACE index and Chao1 index were 3 415.51 and 2 246.51, respectively, which indicated that the diversity index of 873 OTU units showed that the sample library coverage was 0.936 and the ACE index and Chao1 index were 3 415.51 and 2 246.51, respectively, which indicated that there were more microbes and microbes in granular sludge. The group consists of four main categories, Paludibacter paludibacterium Parabacteroidesus Erysipelotrichaceaeae Clostridium phascolarctoibacterium Saccharofermentans and Alkaliflexus (the sum is 24.93333N); hydroacetic acidogenic Petrimonas and trophSynomonas (34.89%; methanosaeta3.44); and Levilina Longilinea and Thervirgaea (the proportion is 14.62g).
