人工光源補(bǔ)光對(duì)沉水植物凈化染料廢水效果及其生理影響研究

【摘要】:沉水植物廣泛應(yīng)用于污染水體的生態(tài)修復(fù),對(duì)水質(zhì)具有良好的凈化作用。針對(duì)染料廢水常規(guī)物化和生化處理方法的不足以及沉水植物應(yīng)用于染料廢水凈化存在的水下光照不足問(wèn)題,提出了人工光源輔助沉水植物凈化染料廢水的新方法,通過(guò)調(diào)控人工光源的光質(zhì)、光強(qiáng)和日補(bǔ)光時(shí)間等參數(shù),改善沉水植物的生境,提高沉水植物的凈化能力,為進(jìn)一步拓寬沉水植物的應(yīng)用和發(fā)展染料廢水治理新技術(shù)提供依據(jù)。論文系統(tǒng)研究了人工光源補(bǔ)光對(duì)典型沉水植物凈化染料廢水效果和生理變化的影響,主要研究?jī)?nèi)容和研究結(jié)果如下:(1)實(shí)驗(yàn)研究了紅、藍(lán)、白三種光質(zhì)人工光源補(bǔ)光對(duì)典型沉水植物金魚(yú)藻、苦草和菹草凈化染料廢水效果的影響,結(jié)果表明,三種光質(zhì)的人工光源補(bǔ)光均可以提高金魚(yú)藻、苦草和菹草的凈化效果;紅光對(duì)金魚(yú)藻和苦草的污染凈化能力的提高效果最佳,藍(lán)光其次,白光最弱;而對(duì)于菹草,藍(lán)光效果最好,紅光次之,白光最弱;紅光補(bǔ)光條件下的金魚(yú)藻污染凈化能力最強(qiáng),停留時(shí)間5d時(shí),可將廢水的色度、總磷、氨氮由初始的40倍、1.5mg/L、8.0mg/L分別降至7倍、0.11mg/L、1.84mg/L,去除率分別可達(dá)到82.5%、92.7%、77.0%。(2)實(shí)驗(yàn)研究了光強(qiáng)和日補(bǔ)光時(shí)間對(duì)金魚(yú)藻凈化能力的影響,結(jié)果表明,實(shí)驗(yàn)范圍內(nèi),光強(qiáng)的增加和日補(bǔ)光時(shí)間的延長(zhǎng)與金魚(yú)藻的污染凈化能力呈正相關(guān),然而考慮經(jīng)濟(jì)因素,適宜的工藝參數(shù)為:功率9w、補(bǔ)光3h、停留時(shí)間5d。(3)以小試確定的適宜工藝參數(shù)(紅光、功率9w、補(bǔ)光3h),對(duì)某園區(qū)的染料生化尾水進(jìn)行中試研究,結(jié)果表明,停留時(shí)間5d,染料生化尾水的色度、TP、氨氮、苯胺和COD的濃度可降至5倍、0.18mg/L、2.96mg/L、0.47mg/L和30.0mg/L,出水達(dá)到《城鎮(zhèn)污水處理廠污染物排放標(biāo)準(zhǔn)》(GB18918—2002)的一級(jí)A標(biāo)準(zhǔn),去除率分別可達(dá)到91.7%、90.3%、76.5%、83.5%和37.5%,處理效果穩(wěn)定,說(shuō)明人工光源輔助沉水植物法是一種有效的染料廢水處理技術(shù),值得推廣。(4)實(shí)驗(yàn)研究了紅、藍(lán)、白三種光質(zhì)人工光源補(bǔ)光對(duì)金魚(yú)藻的生理影響,結(jié)果表明,三種光質(zhì)人工光源均可促使金魚(yú)藻的SOD, CAT和POD酶對(duì)染料脅迫響應(yīng)的提前和酶活性的提高,有利于提高植物抵抗脅迫的能力;其中,紅光光源對(duì)酶活性響應(yīng)的影響尤為突出,可以將CAT和POD的響應(yīng)分別提前4.5d和2d,促進(jìn)生物量的增長(zhǎng),是輔助沉水植物水體凈化的最佳光質(zhì)選擇;實(shí)驗(yàn)范圍內(nèi),光強(qiáng)的增加和日補(bǔ)光時(shí)間的延長(zhǎng)均可以促進(jìn)金魚(yú)藻POD酶響應(yīng)的提前,并且光強(qiáng)的增加和日補(bǔ)光時(shí)間的延長(zhǎng)對(duì)植物MDA和葉綠素的含量基本無(wú)影響。(5)實(shí)驗(yàn)初步考察了金魚(yú)藻提取過(guò)氧化物酶體的穩(wěn)定性和對(duì)染料的降解機(jī)理,結(jié)果表明,金魚(yú)藻提取過(guò)氧化物酶體具有較好的熱穩(wěn)定性、pH穩(wěn)定性和耐藏性(45℃以下、pH=5-7、4℃冷藏6個(gè)月,活性仍能保持50%左右)。并且根據(jù)三維熒光和傅立葉紅外分析結(jié)果,推測(cè)金魚(yú)藻體內(nèi)富含的過(guò)氧化物酶體可以降解大分子難降解的偶氮染料為小分子的芳香胺類物質(zhì)。
[Abstract]:The ecological restoration of submerged macrophytes is widely used in polluted water purification, has good effect on water quality. Due to the shortage of conventional dye wastewater by physicochemical and biochemical treatment method and submerged plants used in dye wastewater purification of underwater light is insufficient, this paper puts forward a new method of dye wastewater purification aided submerged plant artificial light source, through the the regulation of artificial light, light quality, light intensity and light time and other parameters, improve the submerged plant habitat, improve the purification capacity of submerged plants, and provide a basis for the management of the new technology to further broaden the application and development of submerged plant dyestuff wastewater. This dissertation has systematically studied the effect of dye wastewater and physiological changes of artificial light fill purification of typical submerged the plant, the main research contents and results are as follows: (1) experimental study on red, blue and white three kinds of light and artificial light fill of typical submerged plant Physical effects, Ceratophyllum demersum, dye wastewater purification effect of Vallisneria natans and Potamogeton crispus showed that three kinds of artificial light fill light can improve the purification effect of Ceratophyllaceae, Vallisneria natans and Potamogeton crispus; purification ability of hornworts and Vallisneria red pollution and improve the effect of the best, blue, white and blue for the weakest; potamogetoncrispus, had the best effect. Red light, white light, red light under the condition of the weakest; Ceratophyllaceae pollution purification ability was the strongest, the residence time of 5D, the wastewater color, total phosphorus, ammonia nitrogen by 40 times, the initial 1.5mg/L, 8.0mg/L were reduced to 7 times, 0.11mg/L, 1.84mg/L removal rate can reach 82.5%, 92.7%, 77.0%. (3) to determine the optimum scale (red, light, power 9W, 3H) in a pilot study, park dye biochemical tail water in the results show that the residence time of 5D, dye color, biochemical material the tail water of TP, ammonia nitrogen, concentration of aniline and COD can be reduced to 5 times, 0.18mg/L, 2.96mg/L, 0.47mg/L and 30.0mg/L, the effluent can meet the discharge standard of pollutants "urban sewage treatment plant > (GB18918 - 2002) the level of A, the removal rate could reach 91.7%, 90.3%, 76.5%, 83.5% and 37.5%, the treatment effect stable, that artificial light assisted submerged plant is an effective technique of wastewater treatment, it is worthy of promotion. (4) experimental study on the effect of red, blue, white, three kinds of light physical artificial light to fill ceratophyllumdemersum. The results showed that three kinds of light and artificial light can promote the hornwort SOD, C AT and POD enzyme of stress response in advance and enzyme activity of dyes increased, is conducive to the improvement of plant resistance to stress; the red light effect on enzyme activity in response is particularly prominent, can be CAT and POD respectively in response to 4.5d and 2D, to promote the growth of biomass is the best light quality auxiliary submerged plant water purifying selection; experiment range, the increase of light intensity and light on the extension of time all can promote the early response of the hornwort POD enzyme, and the increase of light intensity and prolong the time to fill the basic content of plant chlorophyll MDA and shadowless ring. (5) the preliminary experimental investigation of stability of extraction and peroxisome hornwort dye the results show that the degradation mechanism, extraction of hornwort peroxisome has good thermal stability, pH stability and storability (below 45 DEG pH=5-7,4 DEG C, stored for 6 months, still can maintain activity About 50%). And according to the three-dimensional fluorescence and Fu Liye infrared analysis results, presumably in the body of peroxisomes can degrade hornwort macromolecules difficult degradation of azo dyes for aromatic amine molecules.
