表面連接與核-殼型鍵合FITC熒光納米硅球的制備及性能比較

【摘要】:以異硫氰酸熒光素(FITC)為熒光分子,二氧化硅為生物相容性載體制備熒光納米硅球.為增強(qiáng)硅球的穩(wěn)定性,引入γ-氨丙基三乙氧基硅烷(APTES)做橋梁分子,由于FITC、SiO_2與APTES連接順序的非單一性,分別制備了表面連接型和核-殼型鍵合FITC的熒光硅球,并對(duì)其形貌和光漂白性進(jìn)行了比較.實(shí)驗(yàn)結(jié)果表明,核-殼型和表面連接型熒光硅球形貌相似粒徑接近,但表面連接型光漂白性嚴(yán)重.進(jìn)而,對(duì)核-殼型中典型的包硅方法——St9ber法和反相微乳液法進(jìn)行比較,結(jié)果表明,反相微乳液法制備的核-殼型鍵合FITC的熒光硅球熒光信號(hào)更強(qiáng)、粒徑更小且分散性良好,同時(shí)在7h內(nèi)熒光染料泄漏程度小于8%,30min內(nèi)熒光強(qiáng)度僅降低2.2%,具備低染料泄露、光穩(wěn)定性好等優(yōu)點(diǎn).
[Abstract]:Fluorescent nanocrystals were prepared using fluorescein isothiocyanate (FITC) as fluorescent molecule and silica as biocompatibility carrier. In order to enhance the stability of silica spheres, 緯 -aminopropyltriethoxysilane (APTES) was introduced as a bridge molecule. Due to the non-singularity of the bonding sequence between FITCX SiO-2 and APTES, the fluorescent silica spheres bonded with FITC were prepared, respectively. The morphology and photobleaching properties were compared. The experimental results show that the morphologies of core-shell and surface-connected fluorescent silicon spheres are similar to each other, but the surface connected photolysis is very serious. Furthermore, the typical core-shell silicon inclusion method, St9ber method and inverse microemulsion method, are compared. The results show that the fluorescence signal of the core-shell bonded FITC is stronger, the particle size is smaller and the dispersion is good. At the same time, the leakage degree of fluorescent dye is less than 8% and the fluorescence intensity decreases only 2.2% within 30 min within 7 h, which has the advantages of low dye leakage and good light stability.
