不同BMI的9~12歲男性血清性激素相關(guān)指標(biāo)及脂質(zhì)組學(xué)研究

【摘要】:研究目的:肥胖的男性兒童青少年表現(xiàn)出性激素水平異常的現(xiàn)象。目前,青春期前男性肥胖與性激素水平的研究受到年齡劃分、檢測方法、種族差異等的影響而存在一些爭議。由于脂代謝常規(guī)指標(biāo)(血脂等)、脂質(zhì)代謝產(chǎn)物與性激素指標(biāo)之間均表現(xiàn)出顯著的相關(guān)關(guān)系,因此本研究期望通過了解不同BMI的9~12歲男性的性激素相關(guān)指標(biāo)的差異后,再結(jié)合脂質(zhì)組學(xué)來分析9-12歲男性的脂質(zhì)代謝與性激素相關(guān)指標(biāo)的關(guān)系,為兒童青少年肥胖與性激素的研究提供新的思路。研究內(nèi)容:研究一將了解不同BMI的9~12歲男性脂代謝、性激素相關(guān)指標(biāo)之間的差異。研究二將首次對不同BMI的9~12男性進(jìn)行血清脂質(zhì)組學(xué)研究,篩選差異脂質(zhì)代謝物并從脂質(zhì)組學(xué)角度分析男性肥胖與性激素相關(guān)指標(biāo)的關(guān)系。研究對象和方法:平均年齡為10.28±0.69歲的72名9~12歲男性被納入本論文中并根據(jù)BMI分為正常組(NC組)、超重(ow組)、肥胖組(OB組)。研究一身體發(fā)育指標(biāo)測試按各標(biāo)準(zhǔn)及儀器規(guī)范進(jìn)行;血清指標(biāo)測試采用化學(xué)發(fā)光免疫分析、酶聯(lián)免疫吸附試驗進(jìn)行;數(shù)據(jù)處理采用SPSS22.0軟件進(jìn)行分析。研究二采用Waters UPLC/Q-TOF/MS建立脂質(zhì)組學(xué)分析方法:數(shù)據(jù)采集及差異代謝物鑒定利用MassLynx 4.1軟件、Progenesis QI軟件、simca-p軟件等共同進(jìn)行分析。研究結(jié)果:(1)研究一中Ow組表現(xiàn)為HDL-c降低(p0.01)、性激素結(jié)合球蛋白(SHBG)降低(p0.01)、脫氫表雄酮(DHEA)升高(p0.01)的現(xiàn)象,OB組表現(xiàn)為TG升高(p0.01)、HDL-c降低(p0.01)、SHBG降低(p0.01)、DHEA升高(p0.05)的現(xiàn)象。(2)研究二利用UPLC/QTOF/MS儀器建立了不同BMI的9~12歲男性血清脂質(zhì)組學(xué)方法并篩選、鑒定出153個差異脂質(zhì)代謝物.結(jié)論:(1)超重、肥胖的9~12歲男性其性激素相關(guān)指標(biāo)異常的現(xiàn)象表現(xiàn)為DHEA的上升和SHBG的下降。(2)首次對不同BMI的9~12歲男性進(jìn)行了脂質(zhì)組學(xué)的研究,其中超重肥胖組的脂質(zhì)代謝紊亂現(xiàn)象主要表現(xiàn)為甘油酯類代謝物的升高、甘油磷脂類和鞘脂類代謝物的下降,這為肥胖及肥胖相關(guān)疾病的研究提供了實驗依據(jù)。
[Abstract]:Objective: obese male children and adolescents showed abnormal sex hormone levels. At present, the study of male obesity and sex hormone level before puberty is influenced by age division, detection method, race difference and so on. Since there is a significant correlation between lipid metabolites and sex hormone indices (lipid, etc.), this study is expected to understand the differences of sex hormone related indexes among 9- and 12-year-old men with different BMI. The relationship between lipid metabolism and sex hormone related indexes in men aged 9-12 years was analyzed in order to provide a new idea for the study of obesity and sex hormone in children and adolescents. Content: the first study will understand the differences of lipid metabolism and sex hormone related indexes among 9-year-old men with different BMI. In the second study, the serum lipids of 9 / 12 men with different BMI were studied for the first time, and the relationship between male obesity and sex hormone related indexes was analyzed from the point of view of lipid group. Participants and methods: 72 912 year old men with an average age of 10.28 鹵0.69 years were included in this study and were divided into normal group (NC group), overweight group (ow group) and obese group (OB group) according to BMI. A body development index test was carried out according to each standard and instrument standard; the serum index test was carried out by chemiluminescence immunoassay and enzyme-linked immunosorbent assay (Elisa); and the data processing was analyzed by SPSS22.0 software. In the second study, Waters UPLC/Q-TOF/MS was used to establish the method of lipid analysis: data acquisition and differential metabolites identification were analyzed by MassLynx 4.1 software Progenesis QI software and simca-p software. The results were as follows: (1) in the first Ow group, the HDL-c decreased (p0.01), the sex hormone binding globulin (SHBG) decreased (p0.01), and the dehydroepiandrosterone (DHEA) increased (p0.01). In OB group, the increase of TG (p0.01) and the decrease of HDL-c (p0.01) decreased (p0.01). UPLC/QTOF/MS instrument was used to establish and screen serum lipids of 9- and 12-year-old men with different BMI. 153 different lipid metabolites were identified. Conclusion: (1) the abnormal sex hormone related indexes of 9 ~ 12 years old men who are overweight and obese show that the increase of DHEA and the decrease of SHBG. (2) the lipigraphy of 912 year old men with different BMI was studied for the first time. The disorder of lipid metabolism in overweight and obesity group was mainly characterized by the increase of glycerol ester metabolites, the decrease of glycerol phospholipid and sphingomyelin metabolites, which provided the experimental basis for the study of obesity and obesity related diseases.
