有氧運動對MI后大鼠心肌組織抑炎因子的影響及調(diào)控研究

【摘要】:研究目的:探討有氧運動對心肌梗死(MI)后大鼠心肌組織抑炎因子白介素-4(IL-4)、白介素-10(IL-10)分泌情況及信號傳導蛋白和轉(zhuǎn)錄激活物6(STAT6)磷酸化水平的影響,以及STAT6在炎癥反應中的作用機制。研究方法:選用30只雄性8周齡的SD大鼠,隨機分為假手術(shù)組(Sham)、心肌梗死組(MI)、梗死+運動組(ME)。以冠狀動脈左前降支(LAD)結(jié)扎術(shù)構(gòu)建MI模型,運動方式為間歇有氧運動。運動方案:1周的適應性運動后,10m/min熱身10min,然后以20m/min運動8min,再以15 m/min運動2 min,交替4個循環(huán),最后20 m/min運動8min,10 m/min運動2 min后結(jié)束。每天訓練1次,5 d/w,共訓練8 w。以二維超聲心動圖檢測大鼠心臟室壁厚度、射血功能;采用ELISA的方法檢測大鼠心肌組織IL-4、IL-10的分泌水平;Western blot檢測心肌組織p-STAT6、t-STAT6的表達水平。研究結(jié)果:(1)MI手術(shù)使大鼠心臟心室壁變薄,射血功能降低,射血分數(shù)(EF)與Sham組相比明顯減小(P≤0.01);8周有氧運動使MI大鼠的室壁厚度增加、射血功能加強,ME組舒張末期左室后壁厚度(LVPWd)與舒張末期室間隔厚度之和明顯高于MI組(P≤0.05),ME組EF比MI組明顯升高(P≤0.05)。(2)MI組心重/體重比值(HW/BW)顯著低于Sham組(P≤0.05),ME組與MI組相比有明顯的增加(P≤0.05);而ME組與MI組肺重/體重比值(LW/BW)相比有明顯的降低(P≤0.01)。(3)ME的心肌組織IL-4分泌水平與MI組相比有明顯升高(P≤0.05);ME組心肌組織的IL-10分泌水平也顯著高于MI組(P≤0.05)。(4)Western blot 結(jié)果顯示,ME 組的 p-STAT6/t-STAT6 比值顯著高于MI 組(P≤0.05)。研究結(jié)論:(1)有氧運動有利于MI大鼠心臟舒縮功能的改善;(2)有氧運動有助于MI大鼠心功能的恢復,減輕炎癥反應;(3)有氧運動可以升高MI大鼠心肌組織中抑炎因子IL-4、IL-10的分泌水平;(4)有氧運動有助于激活大鼠心肌組織STAT6的磷酸化改變,從而抑制炎癥反應。
[Abstract]:Objective: to investigate the effects of aerobic exercise on the secretion of interleukin-4 (IL-4), interleukin-10 (IL-10) and phosphorylation of signal transduction protein and activator 6 (STAT6) in myocardial tissue after myocardial infarction (MI) in rats. And the mechanism of STAT 6 in inflammatory reaction. Methods: thirty male 8-week-old SD rats were randomly divided into sham-operated group, myocardial infarction group and myocardial infarction group. The MI model was established by left anterior descending branch ligation of coronary artery. Exercise program: 1 week of adaptive exercise followed by 10 m / min warm up for 10 minutes, then 20m/min for 8 minutes, then 15 m/min for 2 minutes, alternating 4 cycles, and finally 20 m/min for 8 minutes and 10 m/min for 2 min. Practice once a day for 5 d / w, and train for 8 ws. The myocardial wall thickness and ejection function were measured by two-dimensional echocardiography, the level of IL-4 IL-10 was detected by Elisa and the expression of p-STAT6t-STAT6 was detected by Western blot.
