【摘要】：研究不同訓(xùn)練狀態(tài)大鼠心肌糖皮質(zhì)激素受體(GR)和鹽皮質(zhì)激素受體(MR)的表達(dá)規(guī)律,探討其在運(yùn)動(dòng)心肌適應(yīng)過(guò)程中可能產(chǎn)生的作用。方法:大鼠分別采用一次耐力運(yùn)動(dòng)、4周和10周的遞增負(fù)荷耐力運(yùn)動(dòng)訓(xùn)練方案,并在末次運(yùn)動(dòng)后0 h、3 h、24 h取材,采用Western blot方法測(cè)定左室心肌相應(yīng)的指標(biāo)。結(jié)果:腎上腺重量以及腎上腺/體重在4周和10周的訓(xùn)練大鼠均顯著增加;而心肌GR和MR的表達(dá)在4周遞增負(fù)荷耐力訓(xùn)練后與安靜對(duì)照組比較顯著下降,經(jīng)過(guò)10周遞增負(fù)荷耐力訓(xùn)練后,受體表達(dá)量與安靜對(duì)照組無(wú)明顯差異。一次、4周和10周的耐力運(yùn)動(dòng)對(duì)大鼠心肌p-SGK1/SGK1沒(méi)有明顯影響;但心肌HSP70的表達(dá)量隨著運(yùn)動(dòng)訓(xùn)練的持續(xù),呈遞增性提高。結(jié)論:大鼠心肌GR和MR表達(dá)以及受體與配體的親和力與運(yùn)動(dòng)訓(xùn)練狀態(tài)有關(guān),這些變化可能參與了運(yùn)動(dòng)心肌的適應(yīng)過(guò)程。
[Abstract]:To study the expression of glucocorticoid receptor (GRG) and salt corticosteroid receptor (MRR) in rat myocardium under different training conditions and to explore its possible role in the process of exercise myocardial adaptation. Methods: the rats were trained with one endurance exercise for 4 weeks and 10 weeks respectively. The left ventricular myocardial indexes were measured by Western blot method at 0 h, 3 h and 24 h after the last exercise. Results: the weight of adrenal gland and adrenal / body weight increased significantly at 4 and 10 weeks, while the expression of gr and Mr decreased significantly after 4 weeks of incremental stress endurance training. After 10 weeks of incremental load endurance training, there was no significant difference in the expression of receptor between the control group and the quiet control group. Endurance exercise for 4 and 10 weeks had no significant effect on myocardial p-SGK1/SGK1 in rats, but the expression of myocardial HSP70 increased progressively with the continuous exercise training. Conclusion: the expression of gr and Mr and the affinity between receptor and ligand are related to the state of exercise training. These changes may be involved in the adaptation of exercise myocardium.
