急性耐力運(yùn)動(dòng)使心肌氧化應(yīng)激-自噬通量-凋亡信號(hào)向穩(wěn)態(tài)平衡的變化趨勢(shì)調(diào)整

【摘要】:探討交通相關(guān)微粒(diesel exhaust particles,DEP)懸液急性滴注對(duì)心肌氧化應(yīng)激、細(xì)胞自噬和凋亡信號(hào)的影響及急性耐力運(yùn)動(dòng)的調(diào)節(jié)功能與干預(yù)作用。方法:8周健康雄性C57BL/6小鼠,隨機(jī)分為對(duì)照組(Con)、急性耐力運(yùn)動(dòng)組(AE)、急性染毒組(DEP)和急性耐力運(yùn)動(dòng)+染毒組(AE+DEP),每組均6只。DEP組和AE+DEP組進(jìn)行一次急性DEP染毒滴注(1 mg/kg)。隨后,AE組和AE+DEP組進(jìn)行一次75%VO_(2max)的急性耐力跑臺(tái)運(yùn)動(dòng)(12 m/min,90 min)。運(yùn)動(dòng)后恢復(fù)12 h后斷頸椎處死,摘取心臟。酶標(biāo)儀測(cè)心肌氧化應(yīng)激水平(MDA、SOD、CAT和H_2O_2等),Western blotting測(cè)心肌自噬相關(guān)蛋白(Atg5、LC3、P62、Beclin1和UVRAG等)和凋亡相關(guān)蛋白(Caspase 3和PARP等)表達(dá)水平。結(jié)果:1)DEP可使心肌MDA和H_2O_2含量顯著升高,CAT活性明顯下降,而急性耐力運(yùn)動(dòng)可顯著抑制DEP介導(dǎo)的心肌MDA和H_2O_2含量升高,并抑制CAT活性下降;2)DEP可顯著上調(diào)心肌自噬蛋白Atg5、LC3-Ⅱ表達(dá)和LC3-Ⅱ/LC3-Ⅰ比率,降低P62蛋白表達(dá),但Beclin1通路自噬關(guān)鍵蛋白Beclin1和UVRAG表達(dá)卻無(wú)顯著變化,而急性耐力運(yùn)動(dòng)可顯著抑制DEP介導(dǎo)的心肌Atg5和LC3-II蛋白表達(dá)升高、LC3-Ⅱ/LC3-Ⅰ比率上調(diào),并抑制P62蛋白表達(dá)下降,但對(duì)Beclin1和UVRAG蛋白表達(dá)未造成顯著影響;3)DEP可顯著上調(diào)心肌凋亡相關(guān)蛋白Caspase 3和PARP的表達(dá),而急性耐力運(yùn)動(dòng)可抑制心肌Caspase3蛋白過(guò)量表達(dá),并顯著降低PAR P蛋白表達(dá)。結(jié)論:DEP可使心肌產(chǎn)生明顯的氧化應(yīng)激反應(yīng),致使心肌自噬通量水平過(guò)量增加,進(jìn)而誘導(dǎo)心肌細(xì)胞凋亡。急性耐力運(yùn)動(dòng)可下調(diào)心肌氧化應(yīng)激水平,抑制DEP介導(dǎo)的心肌自噬過(guò)度激活和細(xì)胞凋亡。提示:急性耐力運(yùn)動(dòng)使心肌氧化應(yīng)激-自噬通量-凋亡信號(hào)具有向穩(wěn)態(tài)平衡方向發(fā)展的變化趨勢(shì)。
[Abstract]:To investigate the effects of acute infusion of diesel exhaust particlesl (DEP) suspension on myocardial oxidative stress, autophagy and apoptotic signal, and the regulatory function and intervention effect of acute endurance exercise. Methods healthy male C57BL/6 mice were randomly divided into control group, acute endurance exercise group, acute endurance exercise group and acute endurance exercise group. Then the AE group and AE DEP group were given an acute endurance treadmill exercise of 12 m / min / min ~ 90 min ~ (-1) and 75 m 路min ~ (2) 路min ~ (-1) 路min ~ (-1) 路min ~ (-1) respectively. After 12 hours of exercise, the cervical vertebrae was amputated and the heart was removed. The level of oxidative stress in myocardium was measured by enzyme scale. Results the content of MDA and H_2O_2 in myocardium was significantly increased and the activity of catalase was significantly decreased, while acute endurance exercise could significantly inhibit the increase of MDA and H_2O_2 in myocardium mediated by DEP.
