有氧運(yùn)動(dòng)對(duì)MI后大鼠心肌細(xì)胞端粒酶活性及細(xì)胞增殖的影響研究

【摘要】:研究目的:探討有氧運(yùn)動(dòng)對(duì)心肌梗死(MI)后大鼠心肌細(xì)胞端粒酶活性及心肌細(xì)胞增殖的影響。研究方法:選用雄性SD大鼠30只,隨機(jī)分為假手術(shù)組(sham),心肌梗死組(MI),心肌梗死運(yùn)動(dòng)組(ME),每組10只。采用冠狀動(dòng)脈左前降支結(jié)扎手術(shù)建立心肌梗死模型。sham組除不進(jìn)行結(jié)扎外其余手術(shù)過(guò)程同其他組。ME組心梗手術(shù)1周后進(jìn)行為期8周間歇有氧運(yùn)動(dòng)。實(shí)驗(yàn)結(jié)束后,采用二維超聲心動(dòng)圖檢測(cè)大鼠心室厚度和射血功能。稱量計(jì)算心重/體重比值(HW/BW)、左心室重量/體重比值(LVW/BW)。采用ELISA檢測(cè)血漿和心肌組織胰島素樣生長(zhǎng)因子-1(IGF-1)的表達(dá)水平。采用RT-qPCR檢測(cè)心肌組織GATA4和Nkx 2.5mRNA水平,衡量心肌細(xì)胞的增殖能力。采用ELISA檢測(cè)心肌端粒酶活性,并采用RT-qPCR檢測(cè)心肌組織端粒酶逆轉(zhuǎn)錄酶(TERT)和自身攜帶的RNA(TERC)的mRNA水平。研究結(jié)果:1.心梗手術(shù)使大鼠心臟心室壁變薄,射血功能降低。與sham組相比,MI組射血分?jǐn)?shù)(EF)明顯降低(P≤0.01),HW/BW 和 LVW/BW 也明顯降低(P≤0.05vs.P≤0.05);8 周間歇有氧運(yùn)動(dòng)后,ME組心臟舒張末期左室后壁厚度與舒張末期室間隔厚度之和(LVPWd+IVSd),EF 值均比 MI 組明顯增高(P≤0.05 vs.P ≤0.05),HW/BW 和 LVW/BW也有明顯升高(P≤ 0.05 vs.P≤ 0.05)。2.間歇有氧運(yùn)動(dòng)使心梗后大鼠心肌細(xì)胞增殖程度被激活。ME組血漿和心肌組織內(nèi)IGF-1的水平比MI組明顯升高(P≤0.05vs.P≤0.05)。與sham組相比,MI組心肌Nkx 2.5的mRNA表達(dá)水平明顯升高(P≤ 0.05);而ME組心肌GATA4和Nkx 2.5 mRNA表達(dá)水平比MI組顯著升高(P≤0.01 vs.P≤0.05)。3.間歇有氧運(yùn)動(dòng)能明顯升高心梗大鼠心肌組織的端粒酶表達(dá)水平。有氧運(yùn)動(dòng)使ME組心肌組織端粒酶水平與MI組相比顯著升高(P ≤ 0.01)。與Sham組相比,MI組心肌TERT mRNA表達(dá)水平明顯增高(P ≤ 0.05);而ME組心肌TERC和TERT的mRNA表達(dá)水平都比MI組顯著增高(P≤0.01vs.P≤0.01)。研究結(jié)論:1.間歇有氧運(yùn)動(dòng)能明顯促進(jìn)MI后大鼠心肌細(xì)胞增殖,改善心功能。2.間歇有氧運(yùn)動(dòng)能激活MI后大鼠心肌細(xì)胞端粒酶活性,進(jìn)而促進(jìn)心肌細(xì)胞增殖。
[Abstract]:Objective: to investigate the effects of aerobic exercise on telomerase activity and cardiomyocyte proliferation after myocardial infarction (MI) in rats. Randomly divided into sham operation group, myocardial infarction group, myocardial infarction group, myocardial infarction exercise group, 10 rats in each group. Coronary artery left anterior descending branch ligation operation was used to establish myocardial infarction model. Sham group except for no ligation of the rest of the operation process with other groups. ME. Group A received intermittent aerobic exercise for 8 weeks after myocardial infarction operation for 1 week. The ventricular thickness and ejection function of rats were measured by two-dimensional echocardiography. The ratio of heart weight to body weight was calculated and the ratio of left ventricular weight to body weight was measured. The expression of insulin-like growth factor-1 IGF-1 in plasma and myocardium was detected by ELISA. The levels of GATA4 and Nkx 2.5mRNA in myocardial tissue were detected by RT-qPCR. To measure the proliferative ability of cardiomyocytes. To measure the activity of cardiac telomerase by ELISA, and to measure the mRNA level of cardiac tissue by RT-qPCR.
