全血標(biāo)本離心前不同保存溫度與時(shí)間對(duì)巨細(xì)胞病毒核酸檢測(cè)結(jié)果的影響

【摘要】:目的探討全血標(biāo)本離心前不同溫度與時(shí)間對(duì)人巨細(xì)胞病毒(human cytomegalovirus,HCMV)核酸檢測(cè)(nucleic acid amplification test,NAT)結(jié)果的影響,以確定離心前放置的安全時(shí)限。方法采集含高、中、低DNA載量的全血。采血后按保存溫度分為4℃、25℃、37℃組,各組放置0、1、6、12、24、48、72 h后進(jìn)行離心,行實(shí)時(shí)熒光定量PCR。結(jié)果高濃度4℃組離心前放置72 h,25℃與37℃組離心前放置24 h,無明顯變化;中濃度4℃組離心前放置24 h,25℃組、37℃組離心前放置12 h,無明顯變化;低濃度4℃組、25℃組離心前放置12 h,37℃組離心前放置6 h,無明顯變化。結(jié)論為保證HCMV NAT結(jié)果的準(zhǔn)確性,全血標(biāo)本最好在離心前4℃保存,并在采血后6 h內(nèi)及時(shí)離心送檢,高溫條件下更應(yīng)注意。
