【摘要】：目的 研究甲狀旁腺即時(shí)自體移植過(guò)程中,甲狀旁腺組織術(shù)中體外臨時(shí)保存的細(xì)胞活性影響因素；以期找到一種最佳的術(shù)中甲狀旁腺體外臨時(shí)保存條件,使甲狀旁腺組織的活性影響降至最低,以進(jìn)一步提高甲狀旁腺自體移植的成功率。 方法 2014年01月至2014年12月,甲狀腺癌手術(shù)中即刻切取12枚因受腫瘤侵犯而需要一并切除的新鮮甲狀旁腺組織,每個(gè)切成7份小組織塊；隨機(jī)單盲分組,1份組織塊直接新鮮固定,作為對(duì)照組,其余6份分別在室溫生理鹽水(22-24℃)和4℃DMEM培養(yǎng)液中保存0.5小時(shí)、1.0小時(shí)和2.0小時(shí)；最后所有組織塊固定切片,行透射電子顯微鏡檢查,觀察細(xì)胞超微結(jié)構(gòu)變化,評(píng)估細(xì)胞活性情況。 結(jié)果 ①新鮮甲狀旁腺組織,在室溫生理鹽水中保存0.5小時(shí)后可見(jiàn)染色質(zhì)固縮,少量線粒體腫脹變圓；保存1.0小時(shí)后可見(jiàn)核溶解、染色質(zhì)凝集、核內(nèi)空泡化,線粒體腫脹變大變圓、空泡樣變性。 ②新鮮甲狀旁腺組織,在4℃DMEM培養(yǎng)液中保存0.5小時(shí)后甲狀旁腺細(xì)胞超微結(jié)構(gòu)未見(jiàn)明顯變化；保存1.0小時(shí)后,可見(jiàn)細(xì)胞核變大變圓,線粒體腫脹,空泡樣變性。 結(jié)論 甲狀旁腺體外臨時(shí)保存的介質(zhì)和保存時(shí)間長(zhǎng)短是甲狀旁腺細(xì)胞活性的影響因素,在甲狀旁腺即時(shí)自體移植過(guò)程中最好采用4℃DMEM等細(xì)胞培養(yǎng)液作為體外臨時(shí)保存溶液,保存時(shí)間不應(yīng)超過(guò)1.0小時(shí),越快行自體移植越好。
[Abstract]:Objective to study the factors affecting the cell viability of parathyroid tissue during the immediate autotransplantation of parathyroid gland in vitro, and to find out the best condition for the preservation of parathyroid gland in vitro. The activity of parathyroid tissue was minimized to further improve the success rate of autotransplantation of parathyroid gland. Methods from January 2014 to December 2014, 12 fresh parathyroid tissues, which needed to be removed simultaneously because of tumor invasion, were removed immediately during the operation of thyroid carcinoma, each of which was cut into 7 small tissue blocks.
