定量PCR檢測HLA-C等位基因表達(dá)方法的建立和初步應(yīng)用

【摘要】:目的:建立實(shí)時定量PCR方法檢測同一個體兩條不同的HLA-C等位基因的表達(dá)水平。方法:構(gòu)建HLAⅠ類等位基因外顯子2和3數(shù)據(jù)庫,設(shè)計(jì)等位基因特異性系列引物,建立等位基因定量PCR方法;在數(shù)據(jù)庫和835例漢族人外周血標(biāo)本中對方法進(jìn)行理論和實(shí)驗(yàn)驗(yàn)證;檢測并比較20對肝癌和癌旁組織標(biāo)本中HLA-C等位基因的表達(dá)水平。結(jié)果:本實(shí)驗(yàn)設(shè)計(jì)的20對引物能檢測23種雜合基因型中不同的HLA-C等位基因,覆蓋漢族人群中出現(xiàn)頻率大于0.96%的個體;55%(11/20)的肝癌組織中HLA-C位點(diǎn)兩條等位基因表達(dá)水平變化不一致。結(jié)論:成功建立了針對漢族人群的HLA-C等位基因表達(dá)水平的檢測方法;肝癌組織中普遍存在著兩條HLA-C等位基因表達(dá)水平變化不一致的現(xiàn)象。
[Abstract]:Objective: to establish a real-time quantitative PCR method to detect the expression of two different HLA-C alleles in the same body. Methods: HLA class I allele exons 2 and 3 databases were constructed, allele specific series primers were designed and allele quantitative PCR method was established, and the method was verified theoretically and experimentally in the database and 835 peripheral blood samples of Han nationality. The expression level of HLA-C allele in 20 pairs of HCC and adjacent tissues was detected and compared. Results: the 20 pairs of primers designed in this experiment can detect different HLA-C alleles in 23 heterozygous genotypes, and the two alleles of HLA-C locus in HCC tissues with frequency of more than 0.96% and 0.96% were not consistent with each other. Conclusion: a method of detecting HLA-C allele expression in Han population was successfully established and two HLA-C alleles were found to be inconsistent in HCC tissues.
