奧曲肽對LPS處理的A549細(xì)胞代謝的影響

【摘要】:目的:探討生長抑素衍生物奧曲肽對脂多糖(LPS)誘導(dǎo)A549細(xì)胞代謝組學(xué)變化的影響。方法:體外培養(yǎng)肺泡腺癌上皮細(xì)胞A549,分別經(jīng)LPS和LPS+奧曲肽處理,氣相色譜質(zhì)譜聯(lián)用技術(shù)(gas chromatography/mass spectrometry,GC/MS)和液相色譜質(zhì)譜聯(lián)用技術(shù)(liquid chromatography/mass spectrometry,LC/MS)檢測A549細(xì)胞在不同處理?xiàng)l件下的代謝組變化,對結(jié)果進(jìn)行色譜圖可視化檢查,將相應(yīng)數(shù)據(jù)進(jìn)行主成分分析(PCA),解析其差異表達(dá)代謝物,并構(gòu)建互作網(wǎng)絡(luò)圖。結(jié)果:(1)在基于LC/MS方法的代謝組分析中,發(fā)現(xiàn)不同處理組之間有一定差異,進(jìn)一步采用正交潛變量投影判別分析(OPLS-DA),獲得差異性表達(dá)代謝物。差異代謝物以氨基酸類及磷脂類為主。(2)通過GC/MS技術(shù)分析不同處理A549細(xì)胞的代謝組,獲得差異性表達(dá)代謝物,主要為有機(jī)酸,糖類及氨基酸類代謝產(chǎn)物。(3)構(gòu)建互作網(wǎng)絡(luò)圖,主要涉及糖酵解/糖異生、色氨酸代謝、半乳糖代謝、尿素循環(huán)和檸檬酸代謝,并從中發(fā)現(xiàn)14個(gè)具有標(biāo)志性代謝物作用的關(guān)鍵成分,包括5-羥色胺、吲哚、蘇氨酸、絲氨酸、葡萄糖、苯丙氨酸、乳糖、延胡索酸鹽、4-羥基苯乳酸、冬氨酸鹽、天門冬素、腐胺、脯氨酸和琥珀酸鹽。結(jié)論:(1)代謝組分析發(fā)現(xiàn),在奧曲肽處理LPS刺激的A549細(xì)胞的過程中,有機(jī)酸,糖類、氨基酸類和脂類是其變化的主要成分。(2)構(gòu)建了LPS致A549細(xì)胞炎癥反應(yīng)和奧曲肽干預(yù)作用的差異代謝物互作網(wǎng)絡(luò)圖,涉及5個(gè)代謝路徑和14個(gè)關(guān)鍵代謝組分。
[Abstract]:Aim: to investigate the effect of somatostatin derivative octreotide on the metabolic changes of A549 cells induced by lipopolysaccharide (LPS).Methods: A549 cells were cultured in vitro and treated with LPS and LPS octreotide respectively. The metabolic changes of A549 cells were detected by gas chromatography-mass spectrometry (GC / MS) and liquid chromatography-mass spectrometry (LC-MS).The results were visually checked by chromatogram. The corresponding data were analyzed by principal component analysis (PCA), and their differentially expressed metabolites were analyzed, and the interaction network was constructed.Results (1) in the metabolic group analysis based on LC/MS method, we found that there were some differences among different treatment groups, and further used orthogonal latent variable projection discriminant analysis (OPLS-DAA) to obtain the differentially expressed metabolites.The main metabolites were amino acids and phospholipids. (2) the metabolic groups of A549 cells treated with different treatments were analyzed by GC/MS technique, and the differentially expressed metabolites, mainly organic acids, carbohydrates and amino acids metabolites.It is mainly involved in glycolysis / glycometabolism, tryptophan metabolism, galactose metabolism, urea cycle and citric acid metabolism. Among them, 14 key components with iconic metabolites, including 5-hydroxytryptamine, indole, threonine, serine, are found.Glucose, phenylalanine, lactose, fumarate, 4-hydroxyphenyllactic acid, aspartate, asparagin, putrescine, proline and succinate.Conclusion Metabolism analysis showed that organic acids and carbohydrates were involved in the treatment of LPS stimulated A549 cells by octreotide.Amino acids and lipids were the main components of A549 cell line, and constructed a network of different metabolites in A549 cells induced by LPS and Octreotide intervention, involving 5 metabolic pathways and 14 key metabolic components.
