【摘要】：目的對FACSArray流式細(xì)胞儀檢測P選擇素(P-selectin)、組織型纖溶酶原激活物(t-PA)進(jìn)行方法學(xué)性能評價,并觀察其與冠心病各亞組的相關(guān)性。方法用FACSArray流式細(xì)胞儀檢測系統(tǒng)測定P-selectin、t-PA的精密度、準(zhǔn)確度,對靈敏度、干擾試驗等分析性能進(jìn)行驗證和評價。同時,對患有冠心病的患者血清P-selectin、t-PA濃度進(jìn)行檢測,觀察其血清含量與冠心病的相關(guān)性。結(jié)果 P-selectin、t-PA的批內(nèi)和批間變異系數(shù)分別為4.23%~9.00%、4.63%~11.00%和2.20%~2.90%、6.30%~12.20%;P-selectin、t-PA的相對偏倚分別為2.71%~5.28%和1.23%~4.16%,檢測靈敏度為16.20ng/L和1.21ng/mL。高濃度的三酰甘油、血紅蛋白和膽紅素對P-selectin測定時的干擾率分別為14.65%、13.89%和10.94%,對t-PA測定時的干擾率分別為15.65%、13.23%和12.94%;各冠心病亞組P-selectin的濃度范圍為(54 020.1±3 390.1)~(83 189.8±7 226.3)ng/L,t-PA的濃度范圍為(9.59±2.13)~(28.72±2.15)ng/mL,與對照組相比,差異有統(tǒng)計學(xué)意義(P0.05)。結(jié)論FACSArray流式細(xì)胞儀檢測系統(tǒng)檢測血清P-selectin、t-PA的分析性能符合規(guī)定的性能指標(biāo),能夠滿足實驗室認(rèn)可的需要。
[Abstract]:Objective to evaluate the methodological performance of P-selectinus and tissue plasminogen activator (t-PA) detected by FACSArray flow cytometry, and to observe its correlation with various subgroups of coronary heart disease. Methods the precision of P-selectint-PA was measured by FACSArray flow cytometry. Accuracy, sensitivity, interference test, and so on were verified and evaluated. Serum P-selectint-PA levels were also measured in patients with coronary heart disease. Results the coefficients of variation of P-selectint-PA were 4.23ng / ml and 1.23ng/ mL, respectively. The relative bias of P-selectint-PA was 2.715.28% and 1.23ng / mL, respectively. The relative bias of P-selectint-PA was 16.20 ng / L and 1.21 ng / mL, respectively, and the relative bias of P-selectint-PA was 2.715.28% and 1.234.16.The sensitivity was 16.20 ng / L and 1.21 ng / mL respectively, and the relative bias of P-selectint-PA was 2.715.28% and 1.234.16.The sensitivity was 16.20 ng / L and 1.21 ng / mL, respectively. The interference rates of hemoglobin and bilirubin on P-selectin were 13.89% and 10.94, respectively, and the interference rates on t-PA were 15.65 ~ 13.23% and 12.94 respectively. The concentration range of P-selectin in each coronary heart disease subgroup was 54 020.1 鹵3 390.1 ~ 83 189.8 鹵7 226.3 ng / L ~ (t-PA) in the range of 9.59 鹵2.13 ~ (-1) 鹵2.15 ng 路mL ~ (-1), compared with the control group. Conclusion the analytical performance of serum P-selectint-PA by FACSArray flow cytometry can meet the requirements of laboratory accreditation.
