離子液體選擇性萃取分離磷脂酰絲氨酸和磷脂酰膽堿

【摘要】:同系物分離是高純度藥物單體和化學(xué)品制備中的重要環(huán)節(jié),磷脂酰絲氨酸是一類重要的藥物和功能添加劑,其制備過(guò)程中的關(guān)鍵技術(shù)是磷脂酰絲氨酸與其他磷脂同系物的分離,但同系物之間結(jié)構(gòu)相似、分離難度大。報(bào)道了以離子液體為介質(zhì)選擇性萃取分離磷脂酰絲氨酸和磷脂酰膽堿的方法,系統(tǒng)研究了離子液體結(jié)構(gòu)和濃度、萃取溫度對(duì)分離性能的影響。研究表明,離子液體的陰離子結(jié)構(gòu)顯著影響同系物的分離選擇性,且離子液體與極性稀釋劑之間存在協(xié)同萃取效應(yīng),離子液體的摩爾濃度僅為5%時(shí),1-乙基-3-甲基咪唑溴/甲醇-正己烷兩相體系中的磷脂酰絲氨酸和磷脂酰膽堿的選擇性高達(dá)29.48。采用量子化計(jì)算初步研究了萃取機(jī)理,結(jié)果表明離子液體與磷脂之間形成了多重氫鍵和靜電相互作用,從而促進(jìn)了磷脂同系物之間的選擇性分離。
[Abstract]:Homologous separation is an important step in the preparation of high purity drug monomers and chemicals. Phosphatidyl serine is an important class of drugs and functional additives. The key technology in the preparation process is the separation of phosphatidylserine from other phospholipid homologues. However, the structure of homologues is similar and the separation is difficult. The method of selective extraction of phosphatidyl serine and phosphatidylcholine using ionic liquid as the medium was reported. The effects of ionic liquid structure and concentration and extraction temperature on the separation performance were systematically studied. The results showed that the anionic structure of ionic liquids significantly affected the separation selectivity of homologues, and there was a synergistic extraction effect between ionic liquids and polar diluents. The selectivity of phosphatidyl serine and phosphatidylcholine in 1-ethyl-3-methyl imidazolium / methanol-n-hexane two-phase system was 29.48 when the molar concentration of ionic liquid was only 5. The extraction mechanism was preliminarily studied by quantization calculation. The results showed that the ionic liquid and phospholipids formed multiple hydrogen bonds and electrostatic interactions, which promoted the selective separation of phospholipid homologues.
