【摘要】：為了提高多粘菌素E的效價,提出了一種多粘菌素E的生產(chǎn)工藝,應(yīng)用發(fā)酵、膜分離與泡沫分離耦合通過蠕動泵構(gòu)成循環(huán)回流裝置生產(chǎn)多粘菌素E。此工藝發(fā)酵生產(chǎn)多粘菌素E,可以延長菌體生長的穩(wěn)定期,有效解除產(chǎn)物抑制,防止菌體被泡沫帶出,保證營養(yǎng)物質(zhì)充分利用。研究了不同耦合開始時間、間歇時間、流速、單次分離時間對多粘菌素E效價的影響。結(jié)果表明,設(shè)定在23.0 h開始耦合,間歇時間為1.0 h,液體流速為20 m L×min~(-1),單次分離時間為45 min的條件下,發(fā)酵48.0 h多粘菌素E的效價達(dá)到28634 U×m L~(-1),相比非耦合發(fā)酵多粘菌素E的效價(23597U×m L~(-1))提高了21%�？偟膩碚f,該工藝有效地提高了多粘菌素E效價,對于其他表面活性物質(zhì)的分離也有重要的意義。
[Abstract]:In order to improve the titer of polymyxin E, a production process of polymyxin E was put forward. By using fermentation, membrane separation and foam separation, polymyxin E was produced by a circulating reflux device composed of peristaltic pump.The production of polymyxin E by this process can prolong the stable period of cell growth, effectively remove the inhibition of the product, prevent the bacteria from being carried out by foam, and ensure the full utilization of nutrients.The effects of different coupling start time, intermittent time, flow rate and single separation time on the titer of polymyxin E were studied.The results show that when the coupling is started at 23.0 h, the intermittent time is 1.0 h, the liquid flow rate is 20 mL 脳 min-1, the single separation time is 45 min.The titer of polymyxin E reached 28634 U 脳 mL ~ (-1) after 48.0h fermentation, which was 21% higher than that of uncoupled fermentation (23597U 脳 mL ~ (-1)).In general, this process can effectively improve the titer of polymyxin E, and it is also of great significance for the separation of other surface active substances.
