漳平市杜鵑花產(chǎn)業(yè)調(diào)查與VIGS技術(shù)體系初步構(gòu)建

【摘要】:本研究通過(guò)文獻(xiàn)檢索、實(shí)地走訪和電話咨詢(xún)等方式進(jìn)行漳平市杜鵑花產(chǎn)業(yè)現(xiàn)狀分析,從中發(fā)現(xiàn)杜鵑產(chǎn)業(yè)存在的問(wèn)題,最后提出相應(yīng)對(duì)策,旨在促進(jìn)漳平市杜鵑花產(chǎn)業(yè)健康持續(xù)發(fā)展。目前漳平市杜鵑花產(chǎn)業(yè)存在品種引種為主,缺乏自主培育品種問(wèn)題,同時(shí)杜鵑花多以雜交育種為主,所需時(shí)間長(zhǎng),品種更新緩慢,而VIGS技術(shù)作為一種簡(jiǎn)單、快速、高通量的基因功能分析方法,還未應(yīng)用到杜鵑中,因此本研究開(kāi)展了杜鵑花VIGS技術(shù)的體系構(gòu)建,進(jìn)行杜鵑花觀賞性相關(guān)基因的功能驗(yàn)證,利于漳平市杜鵑花新品種的培育,從而促進(jìn)杜鵑產(chǎn)業(yè)發(fā)展。本文的主要研究結(jié)果如下:1.對(duì)漳平市杜鵑花產(chǎn)業(yè)的生產(chǎn)情況、商品情況、發(fā)展優(yōu)勢(shì)和經(jīng)營(yíng)情況進(jìn)行調(diào)查,結(jié)果如下:(1)龍巖作為福建省杜鵑花產(chǎn)業(yè)的主產(chǎn)區(qū),2015年種植面積達(dá)400 hm2,占福建省87%;(2)漳平市杜鵑花產(chǎn)業(yè)規(guī)模雖然有所縮小,甚至在2013年出現(xiàn)產(chǎn)值下滑18%的情況,但是近兩年漳平市專(zhuān)業(yè)化生產(chǎn)水平逐步成熟、生產(chǎn)技術(shù)不斷提高,經(jīng)濟(jì)效益不斷提高,產(chǎn)值呈現(xiàn)上升趨勢(shì)(3)搜集到的漳平市杜鵑花產(chǎn)業(yè)中約100個(gè)杜鵑花品種,從中發(fā)現(xiàn)的杜鵑商品花色主要為紅色和紫色,花形特征為大花和重瓣;盆花和苗木為主要的商品類(lèi)型,盆景杜鵑是新興的商品類(lèi)型(4)漳平市杜鵑花產(chǎn)業(yè)由于土地與租金問(wèn)題,主要以個(gè)體農(nóng)戶(hù)為經(jīng)營(yíng)模式;銷(xiāo)售模式大部分為傳統(tǒng)的批發(fā),開(kāi)店,進(jìn)駐商場(chǎng)的方式,互聯(lián)銷(xiāo)售模式逐漸興起。2.漳平市杜鵑花產(chǎn)業(yè)發(fā)展中存在的問(wèn)題有:(1)商品化品種依賴(lài)引種,缺乏自主知識(shí)產(chǎn)權(quán)品種;(2)產(chǎn)品類(lèi)型和產(chǎn)品應(yīng)用創(chuàng)新能力不足;(3)產(chǎn)業(yè)運(yùn)營(yíng)有待完善,花企缺乏綜合競(jìng)爭(zhēng)力。3.漳平市杜鵑花產(chǎn)業(yè)發(fā)展的優(yōu)勢(shì)條包括:適宜的氣候環(huán)境和獨(dú)特的區(qū)位交通優(yōu)勢(shì),悠久的杜鵑花栽培歷史,政府部門(mén)和相關(guān)政策的支持,豐富的種質(zhì)資源,充沛的勞動(dòng)力,以及良好的花卉市場(chǎng)發(fā)展?jié)摿Α?.通過(guò)對(duì)漳平市杜鵑花產(chǎn)業(yè)存在的問(wèn)題和其優(yōu)勢(shì)條件的分析,提出了一系列有助于漳平市杜鵑花產(chǎn)業(yè)發(fā)展的對(duì)策:(1)重視新品種研發(fā),提升品種創(chuàng)新能力;(2)豐富產(chǎn)品類(lèi)型,拓寬產(chǎn)品應(yīng)用;(3)加強(qiáng)產(chǎn)學(xué)研合作,提升花企綜合競(jìng)爭(zhēng)力;(4)提升產(chǎn)業(yè)現(xiàn)代化水平,優(yōu)化產(chǎn)業(yè)運(yùn)營(yíng)。5.基于煙草脆裂病毒載體,進(jìn)行杜鵑VIGS病毒載體的構(gòu)建。成功克隆了杜鵑392 bp的PDS基因片段、472bp的CHS基因片段、400bp的DFR基因片段、372bp的RDR2A基因片段和350bp的MADS基因片段。同時(shí)構(gòu)建了杜鵑VIGS病毒載體包括,VIGS-PDS 載體、VIGS-CHS 載體、VIGS-DFR 載體、VIGS-MADS載體、VIGS-RDR2A載體。6.杜鵑VIGS技術(shù)體系的建立與優(yōu)化。以杜鵑嫩葉和成熟葉為實(shí)驗(yàn)材料,設(shè)置不同處理的侵染液濃度OD600值分別為1.0、1.5和2.0,采用注射法將含有VIGS-PDS載體的農(nóng)桿菌瞬時(shí)轉(zhuǎn)化杜鵑植株。通過(guò)對(duì)侵染后葉片的表型觀察和統(tǒng)計(jì),發(fā)現(xiàn)杜鵑嫩葉和成熟葉均出現(xiàn)PDS基因表達(dá)受抑制的表型。初步表明采用pTRV病毒載體可以用于杜鵑植株的基因沉默。并且在綜合分析了不同侵染液濃度(OD600值)對(duì)杜鵑葉片PDS基因沉默的效果,以及對(duì)葉片生長(zhǎng)發(fā)育的影響,確定了較適宜的侵染液OD600值為1.0。7.杜鵑花器官VIGS技術(shù)的構(gòu)建與優(yōu)化。設(shè)置OD600值為1.0和2.0兩種侵染液濃度,采用注射法和涂抹法處理杜鵑的花苞和半開(kāi)放的花苞的基部,以及開(kāi)放花的花瓣。對(duì)侵染后的花器官進(jìn)行表型觀察和統(tǒng)計(jì),發(fā)現(xiàn)開(kāi)放花均未出現(xiàn)預(yù)期的CHS基因、DFR基因、MADS基因和RDR2基因表達(dá)受抑制的表型。但通過(guò)比較分析各組實(shí)驗(yàn)處理結(jié)果,本文對(duì)杜鵑花器官VIGS體系的構(gòu)建提出了一些值得參考的建議。
[Abstract]:In this study, the current situation of Rhododendron industry in Zhangping was analyzed through literature search, field visit and telephone consultation, and the existing problems of Rhododendron industry were found. Finally, the corresponding countermeasures were put forward to promote the healthy and sustained development of the azalea industry in Zhangping. At present, the azalea industry in Zhangping was mainly introduced and lack of independent culture. At the same time, the Rhododendron is mostly hybrid breeding, with long time and slow variety, and VIGS technology is not used in Rhododendron as a simple, fast, high throughput gene function analysis method. Therefore, the system of Rhododendron VIGS is constructed and the function of Rhododendron ornamental related genes is tested. It is beneficial to the cultivation of new Rhododendron varieties in Zhangping to promote the development of Rhododendron industry. The main research results of this paper are as follows: 1. the production situation, commodity situation, development advantage and management situation of the azalea industry in Zhangping are investigated. The results are as follows: (1) Longyan is the main producing area of the Rhododendron industry in Fujian Province, and the planting area is 40 in 2015. 0 Hm2, accounting for 87% in Fujian province; (2) although the size of the Rhododendron industry in Zhangping has narrowed, even in 2013, the output value of the city has declined by 18%. But in the last two years, the professional production level of Zhangping is gradually mature, the production technology is increasing, the economic benefit is increasing, and the output value is rising (3) of the Rhododendron industry of Zhangping, which is about 1. 00 Rhododendron varieties, from which Rhododendron is mainly red and purple, flower shape features large flowers and heavy petals, potted flowers and seedlings are the main commodity types, and bonsai Rhododendron is a new commodity type (4) the Rhododendron industry in Zhangping city is mainly based on individual farmers as the management model because of land and rent. The existing problems in the development of.2. Zhangping azalea industry are as follows: (1) commodity variety dependence introduction, lack of independent intellectual property rights variety; (2) product type and product application innovation ability is insufficient; (3) industry operation needs to be improved, flower enterprises lack comprehensive competitiveness. .3. Zhangping Rhododendron industry development advantages include: suitable climate environment and unique regional traffic advantages, long history of Rhododendron cultivation, the support of government departments and related policies, rich germplasm resources, abundant labor force, and good flower market development potential.4. through the existence of the Rhododendron industry in the city of Zhangping The problem and the analysis of its predominance, put forward a series of countermeasures to help the development of Zhangping Rhododendron industry: (1) attach importance to new varieties research and development, improve the ability of variety innovation; (2) enrich product types, broaden the application of products; (3) strengthen the cooperation of production, research and research, improve the comprehensive competitiveness of the enterprises; (4) upgrade the level of industrial modernization and optimize the industrial operation.5. Based on the tobacco embrittlement virus vector, the vector of Rhododendron VIGS virus was constructed. The PDS gene fragment of Rhododendron 392 BP, the CHS gene fragment of 472bp, the DFR fragment of 400bp, the RDR2A gene fragment of 372bp and the MADS gene fragment of 350bp were successfully cloned. The establishment and optimization of R carrier, VIGS-MADS carrier, VIGS-RDR2A vector.6. Rhododendron VIGS technology system, with Rhododendron leaves and mature leaves as experimental materials, the OD600 values of different treatments were 1.0,1.5 and 2 respectively, and the Agrobacterium containing VIGS-PDS carrier was transformed into Rhododendron by injection method. Phenotypic observation and statistics showed that PDS gene expression was inhibited in both young and mature leaves of Rhododendron. It was preliminarily indicated that the use of pTRV virus vector could be used in the gene silencing of Rhododendron plants, and the effect of PDS gene silencing on the leaves of Rhododendron (OD600) and the growth and development of leaves were analyzed in a comprehensive analysis. The optimum OD600 value of the OD600 was to construct and optimize the VIGS technology of the 1.0.7. Rhododendron organ. The concentration of OD600 was 1 and 2, the base of the bud and the semi open bud of Rhododendron and the open flower petals were treated with the injection method and the smear method. The phenotypic observation and statistics of the flower organs after the infection were observed. It was found that the expected CHS gene, the DFR gene, the MADS gene and the RDR2 gene were inhibited in the open flower, but by comparing and analyzing the results of the experimental treatment, some suggestions for the construction of the VIGS system of the azalea organ were put forward.
