【摘要】：生物電子等排體是指一類化合物或基團(tuán),這類物質(zhì)擁有近似的分子形狀和體積,類似的電子分布,并由此表現(xiàn)出相似的物理特性。作為激動(dòng)劑或拮抗劑生物電子等排體能對(duì)相同的生化相關(guān)體系發(fā)揮作用,產(chǎn)生彼此互相相關(guān)的生物特性。藥物結(jié)構(gòu)中酰胺結(jié)構(gòu)作為藥效團(tuán)和藥物的重要組成部分被廣泛應(yīng)用。然而作為藥物分子結(jié)構(gòu)的重要組成部分,酰胺結(jié)構(gòu)也存在著一些明顯的缺點(diǎn),包括代謝不穩(wěn)定、代謝產(chǎn)物有毒和膜滲透性差等。為了克服這些缺點(diǎn)同時(shí)又想保持部分酰胺的特性,生物電子等排體替換是有效的方法之一。在先導(dǎo)化合物的優(yōu)化過(guò)程中,通過(guò)酰胺生物電子等排體替換還能達(dá)到提高生物活性和靶點(diǎn)選擇性、降低合成難度、擴(kuò)展或突破知識(shí)產(chǎn)權(quán)限制等目的。本綜述著重介紹了近五年來(lái)酰胺及其生物電子等排體在先導(dǎo)化合物優(yōu)化過(guò)程中的應(yīng)用。希望能對(duì)含有酰胺結(jié)構(gòu)的先導(dǎo)化合物的優(yōu)化提供新的思路,加速新藥研發(fā)進(jìn)程。
[Abstract]:A bioelectron equivalent is a class of compounds or groups that have similar molecular shapes and volumes, similar electron distributions, And it shows similar physical properties. As agonist or antagonist, bioelectron excretion plays an important role in the same biochemical related system. Producing interrelated biological properties. Amide structures are widely used as pharmacophore and important components of drugs in drug structures. However, as important components of drug molecular structures, The structure of amides also has some obvious disadvantages, including unstable metabolism, toxic metabolites and poor membrane permeability. In the process of optimization of leading compounds, amidebion isobaric substitution can improve biological activity and target selectivity, and reduce the difficulty of synthesis. This review focuses on the application of amides and their bioelectronic isobarkers in the optimization of lead compounds in the past five years. It is hoped that this review can be used for the optimization of lead compounds with amide-containing structures. Optimization provides new ideas, Speed up the development of new drugs.
