KCNQ1內含子1a中STR基因座遺傳多態(tài)性及PIA分型

【摘要】:目的調查漢族群體KCNQ1基因內含子1a中STR基因座的遺傳多態(tài)性,并采用PIA分型技術確定等位基因的親代來源。方法用PCR-STR分型技術對230例武漢漢族無關個體樣本進行KCNQ1基因內含子1a中STR基因座分型檢測;同時選用兩種甲基化敏感的限制酶(msRE)HhaⅠ和HpaⅡ對家系中孩子的基因組DNA進行消化后,采用PIA分型技術檢測父源等位基因。結果KCNQ1內含子1a中STR基因座在漢族人群中檢出10個等位基因、24種基因型,其個體識別能力(PD)、多態(tài)性信息含量(PIC)和非父排除率(PE)分別為0.852、0.66和0.484。HhaⅠ和HpaⅡ可消化個體的母源等位基因,PIA分型僅能檢測出單一的父源等位基因。結論KCNQ1內含子1a中STR基因座在漢族群體具有較高的遺傳多態(tài)性,PIA分型技術可以確定個體等位基因的親代來源,具有較高的法醫(yī)學應用價值。
