牛四核苷酸STR基因座的篩選和多態(tài)性分析

【摘要】:目的篩選陰影帶出現(xiàn)率低且多態(tài)性較高的牛四核苷酸STR基因座。方法用Tandem repeats finder軟件搜索;蚪M中的四核苷酸重復(fù)STR序列片段,用Primer Premier 5.0軟件設(shè)計(jì)引物,然后擴(kuò)增、電泳,篩選出符合要求的STR基因座,并對(duì)100份無關(guān)黃牛個(gè)體血樣進(jìn)行STR基因座分型。結(jié)果共篩選出6個(gè)具有多態(tài)性的牛四核苷酸重復(fù)STR基因座(B006、B007、B008、B022、B025、B026),其100份無關(guān)黃牛個(gè)體血樣的累積個(gè)體識(shí)別率和累積非父排除概率分別為0.99995和0.859591。結(jié)論本研究篩選出的6個(gè)四核苷酸STR基因座陰影帶出現(xiàn)率低且多態(tài)性較高,可用于牛的個(gè)體識(shí)別和親子鑒定的研究。
[Abstract]:Objective to screen bovine tetranucleotide STR loci with low frequency of shadow bands and high polymorphism. Methods Tandem repeats finder software was used to search repeated STR sequences in bovine genome, primers were designed with Primer Premier 5.0 software, then amplified, electrophoresis was used to screen the STR loci which met the requirements. STR locus typing was performed on 100 unrelated bovine blood samples. Results A total of 6 polymorphic STR loci of bovine tetranucleotide repeats (B006B007, B008, B022, B025, B026) were screened. The cumulative individual identification rates and cumulative non-paternal exclusion rates of 100 unrelated bovine blood samples were 0.99995 and 0.8595991, respectively. Conclusion the six tetranucleotide STR loci screened in this study have low occurrence rate and high polymorphism, which can be used in individual identification and paternity identification of cattle.
