【摘要】：目的:探討創(chuàng)傷皮膚組織基質(zhì)金屬蛋白酶(Matrix metalloproteinase-s,MMPs)-2、9的表達(dá)與損傷時(shí)間的相關(guān)性,為損傷時(shí)間推斷提供實(shí)驗(yàn)依據(jù)。方法:建立大鼠皮膚切創(chuàng)模型,應(yīng)用免疫組織化學(xué)技術(shù)、圖像分析技術(shù)檢測(cè)不同損傷時(shí)間大鼠皮膚組織生前及死后MMP-2與MMP-9表達(dá)。結(jié)果:生前傷組大鼠損傷后皮膚組織內(nèi)MMP-2與MMP-9在損傷區(qū)及損傷區(qū)周圍皮膚組織中表達(dá)明顯高于正常對(duì)照組(P0.05)。死后傷組皮膚組織中MMP-2與MMP-9表達(dá)與正常對(duì)照組比較無(wú)明顯差異(P0.05)。生前傷組中MMP-2在損傷后0.5h可見(jiàn)在中性粒細(xì)胞中陽(yáng)性表達(dá),隨時(shí)間進(jìn)展表達(dá)逐漸增強(qiáng),并可見(jiàn)在少量血管內(nèi)皮細(xì)胞中陽(yáng)性表達(dá),12h后明顯增多,3d達(dá)到高峰,維持到第5d后在炎細(xì)胞中表達(dá)顯著減少,而在成纖維細(xì)胞,血管內(nèi)皮細(xì)胞中表達(dá)增加,維持在一個(gè)相對(duì)穩(wěn)定的水平,12～14d恢復(fù)正常。MMP-9在損傷后1h可見(jiàn)在中性粒細(xì)胞中陽(yáng)性表達(dá),隨時(shí)間進(jìn)展表達(dá)逐漸增強(qiáng),12h后明顯增多,3d達(dá)到高峰,維持到第5d,5d后在炎細(xì)胞中表達(dá)顯著減少,而在成纖維細(xì)胞,血管內(nèi)皮細(xì)胞中表達(dá)增加,維持在一個(gè)相對(duì)穩(wěn)定的水平,12～14d恢復(fù)正常。結(jié)論:皮膚損傷組織MMP-2與MMP-9表達(dá)具有規(guī)律性且有良好的時(shí)間相關(guān)性,可望成為法醫(yī)學(xué)損傷時(shí)間推斷的指標(biāo)。
[Abstract]:Aim: to investigate the relationship between the expression of matrix metalloproteinase (Matrix metalloproteinase-s,MMPs)-2 and injury time in traumatic skin and to provide experimental evidence for estimating injury time. Methods: the rat skin injury model was established and the expression of MMP-2 and MMP-9 was detected by immunohistochemistry and image analysis before and after death. Results: the expression of MMP-2 and MMP-9 in the skin tissue of the injured group was significantly higher than that of the normal control group (P0.05). There was no significant difference in the expression of MMP-2 and MMP-9 between postmortem injury group and normal control group (P0.05). Positive expression of MMP-2 was observed in neutrophil at 0.5 h after injury, and increased gradually with time, and increased significantly in a small number of vascular endothelial cells at 12 h after injury. The expression of MMP-9 was significantly decreased in inflammatory cells at the 5th day, but increased in fibroblasts. The positive expression of MMP-9 was found in neutrophils 1 h after injury at a relatively stable level of 12d to 14d, and the positive expression of MMP-9 was found in neutrophilic granulocytes at 1 hour after injury, and the positive expression of MMP-9 was found in neutrophils 1 h after injury. After 12 hours, the expression increased to the peak at the 3rd day, decreased significantly in the inflammatory cells after 5 days, but increased in the fibroblasts and vascular endothelial cells. It was maintained at a relatively stable level and recovered to normal at 12 ~ 14 d. Conclusion: the expression of MMP-2 and MMP-9 in skin injury tissues is regular and has a good time correlation, which is expected to be an index for estimating the time of injury in forensic medicine.
