氯胺酮在急性中毒大鼠體內(nèi)的死后再分布研究

【摘要】:目的探索氯胺酮在大鼠體內(nèi)的死后再分布變化規(guī)律及溫度對再分布的影響。方法48只雄性SD大鼠隨機(jī)分為2個實(shí)驗(yàn)組(室溫組24只、冷藏組18只)和1個對照組(6只),實(shí)驗(yàn)組大鼠以氯胺酮290mg/kg灌胃, 45min后缺氧處死,分別置于室溫(24℃)和冷藏(4℃)條件下,于死后不同時間(0、12、24、48h)取心血、外周血、肝、肺、腎、心肌、大腦,檢測其中氯胺酮含量;對照組大鼠以生理鹽水灌胃,各對應(yīng)組織器官樣品為空白對照。血和組織樣品中加入內(nèi)標(biāo)物SKF_(525A)后堿化,乙酸乙酯萃取,GC/MS全掃描定性,內(nèi)標(biāo)法、工作曲線法氣相色譜定量分析。結(jié)果室溫條件下,大鼠死后48h內(nèi)隨著死亡時間延長,心血、肺、肝中氯胺酮的濃度呈升高趨勢(P0.05),腎臟中氯胺酮的濃度先升高后下降(P0.05),外周血、心肌和腦中氯胺酮的濃度無顯著性變化(P0.05)。冷藏條件下,血液及組織中氯胺酮濃度變化無顯著性差異(P0.05),除心肌外,各樣本濃度均低于相應(yīng)時段室溫條件保存的樣本。結(jié)論氯胺酮在大鼠體內(nèi)存在死后再分布現(xiàn)象。溫度對大鼠死后血液及組織中氯胺酮濃度變化有較明顯的影響。
