【摘要】：目的建立脫落細(xì)胞負(fù)壓吸附方法,用于DNA檢驗中衣物等載體上人體脫落細(xì)胞的采集。方法檢材包括由志愿者佩帶1、5、10、20m in紗線手套,穿著5、10、20m in的內(nèi)衣以及外套、鞋、帽子、頭套、襪子、水杯、礦泉水瓶等。在吸塵器的進(jìn)風(fēng)口處連接特制的吸筒,一端覆蓋具有攔截和靜電吸附細(xì)胞能力的特制吸附膜,選擇適當(dāng)負(fù)壓對各檢材相應(yīng)部位進(jìn)行吸掃。收集吸附膜上的脫落細(xì)胞,采用Chelex-100法提取DNA,AmpFLSTR Identifiler復(fù)合擴(kuò)增試劑盒擴(kuò)增檢測。結(jié)果上述檢材用本文負(fù)壓吸附法采集人體脫落細(xì)胞,經(jīng)檢驗均獲得清晰、完整的STR分型圖譜,并與檢材提供者的基因型一致。結(jié)論本文建立的脫落細(xì)胞負(fù)壓吸附技術(shù)可有效吸附載體上的脫落細(xì)胞,適用于DNA檢案實踐。
[Abstract]:Objective to establish a negative pressure adsorption method for the collection of exfoliated cells on clothing carriers in DNA. Methods the samples were carried out by volunteers, including the wearing of gloves, underwear and jackets, shoes, hats, headsets, socks, water cups, water bottles and so on. A special suction tube is connected at the inlet of the vacuum cleaner, and one end is covered with a special adsorption film with the ability of intercepting and electrostatic adsorption cells, and the appropriate negative pressure is selected to absorb and sweep the corresponding parts of the samples. The exfoliated cells from the adsorbed membrane were collected, and Chelex-100 method was used to extract DNAAmpFLSTR Identifiler and detect the amplification of AmpFLSTR Identifiler. Results the exfoliated cells were collected by the method of negative pressure adsorption in this paper. The STR genotyping patterns were all clear and complete, which were consistent with the genotypes of the sample providers. Conclusion the abscission cell negative pressure adsorption technique established in this paper can effectively adsorb the exfoliated cells on the carrier and can be used in the practice of DNA.
