華東地區(qū)漢族人群7個(gè)Y-STR基因座遺傳多態(tài)性的研究

【摘要】: 目的:調(diào)查5個(gè)新的Y-STR基因座DYS708、DYS712、DYS713、DYS714、DYS715及經(jīng)驗(yàn)證多態(tài)性好的2個(gè)DYS576、DYS709在華東地區(qū)漢族人群中遺傳多態(tài)性,評(píng)估其在法醫(yī)學(xué)中的應(yīng)用價(jià)值;建立DYS708、DYS713、DYS715復(fù)合擴(kuò)增體系。 方法:隨機(jī)篩選108名華東地區(qū)漢族男性個(gè)體,Chelex-100方法提取樣本DNA,利用PCR擴(kuò)增、聚丙烯酰胺凝膠電泳、硝酸銀染色進(jìn)行分型檢測(cè)DYS576、DYS708、DYS709、DYS712、DYS713、DYS714、DYS715基因座的遺傳多態(tài)性。對(duì)7個(gè)基因座等位基因測(cè)序,構(gòu)建等位基因標(biāo)準(zhǔn)物,按照ISFG原則命名等位基因。 結(jié)果:DYS708、DYS712、DYS713、DYS714、DYS715、DYS576、DYS709在該人群中分別檢出了7、13、7、10、7、6和8個(gè)等位基因,其基因多樣性分別為0.7242、0.9115、0.8041、0.8492、0.8121、0.7351和0.8202,這7個(gè)Y-STR基因座的單倍型基因多樣性是0.99999851。測(cè)序結(jié)果顯示:在所選的Y-STR基因座中DYS708是復(fù)合重復(fù)序列,其他是簡(jiǎn)單復(fù)合序列, DYS708、DYS712、DYS713、DYS714、DYS715、DYS576、DYS709的串聯(lián)重復(fù)單位分別為AGAT/AGCA、ATCT、TCTT、CTTTT、AGAT、AAAG和CTTT。建立了銀染復(fù)合擴(kuò)增檢測(cè)3個(gè)Y-STR基因座的分型方法,其分析結(jié)果正確,具有良好的穩(wěn)定性和較好的靈敏性。 結(jié)論:這7個(gè)Y-STR基因座DYS576、DYS708、DYS709、DYS712、DYS713、DYS714、DYS715基因座具有較高的遺傳多態(tài)性,在法醫(yī)學(xué)及人類遺傳學(xué)方面具有較高應(yīng)用價(jià)值。這3個(gè)Y-STR復(fù)合擴(kuò)增體系在建立Y染色體STR數(shù)據(jù)庫(kù)、研究群體遺傳和進(jìn)行法醫(yī)學(xué)鑒定有重要意義。
[Abstract]:Objective: to investigate the genetic polymorphisms of five new Y-STR loci, DYS708, DYS712, DYS713, DYS714, DYS715 and 2 DYS576DYS709, and to evaluate their application value in forensic medicine, and to establish a DYS708DYS713DYS715 multiplex amplification system. Methods: the DNA samples were extracted by Chelex-100 method from 108 Han males in East China. The genetic polymorphisms of DYS576 DYS708 / DYS709 / DYS712DYS713 / DYS714DYS715 locus were detected by PCR amplification, polyacrylamide gel electrophoresis and silver nitrate staining. The alleles of 7 loci were sequenced and the allele standard was constructed. The alleles were named according to ISFG principle. Results DYS708 / DYS712DYS713 / DYS714DYS715 / DYS576DYS709 alleles were detected in this population, respectively. The genetic diversity of the 7 Y-STR loci was 0. 999998551 and 0. 99995851respectively, and the genetic diversity was 0. 72420.91150.8041ng0.84920.8121a 0.7351 and 0. 82022. The genetic diversity of the 7 Y-STR loci was 0. 99999851. the genetic diversity of these alleles was 0. 99999851. the genetic diversity of DYS708, DYS712, DYS713, DYS713 and DYS713 was 0. The sequencing results showed that DYS708 was a complex repeat sequence in the selected Y-STR locus, while the other was a simple complex sequence. The tandem repeat units of DYS708 / DYS712 / DYS713 / DYS715 / DYS576 / DYS709 were AGATR / AGCAATATCTT TCTTTTT AGATA AG and CTT T, respectively. A silver staining multiplex amplification method for the detection of three Y-STR loci was established. The results were correct and had good stability and sensitivity. Conclusion: these seven Y-STR loci DYS576DYS708DYS709 / DYS712DYS713DYS714DYS715 have high genetic polymorphism and have high application value in forensic medicine and human genetics. These three Y-STR multiplex amplification systems are of great significance in the establishment of Y chromosome STR database, the study of population inheritance and forensic identification.
