【摘要】：目的:明確益氣解毒方對(duì)ACHN腎癌干細(xì)胞的增殖及凋亡的影響。方法:采用高、中、低3種濃度的“益氣解毒方”藥物血清干預(yù)體外培養(yǎng)的ACHN腎癌干細(xì)胞。1.采用二甲基偶氮唑藍(lán)(MTT)實(shí)驗(yàn)檢測(cè)“益氣解毒方”藥物血清對(duì)ACHN腎癌干細(xì)胞增殖的影響;2.流式細(xì)胞儀檢測(cè)“益氣解毒方”藥物血清干預(yù)下ACHN腎癌干細(xì)胞的凋亡;3.流式細(xì)胞儀檢測(cè)“益氣解毒方”藥物血清干預(yù)下ACHN腎癌干細(xì)胞P53蛋白的表達(dá)情況。結(jié)果:1.“益氣解毒方”藥物血清可以抑制ACHN腎癌干細(xì)胞的增殖,各組抑制率分別為:空白對(duì)照組24 h:0.36%,48h:0.41%,72:0.43%;低濃度組24 h:15.7%,48h:32.7%,72h:44.3%;中濃度組24h:36.7%,48h:53.3%,72h:63.7%;高濃度組24h:53.3%,48h:67%,72h:77.7%;各組間比較P值均0.05,差異有統(tǒng)計(jì)學(xué)意義。2.三種濃度“益氣解毒方”藥物血清下均可誘導(dǎo)ACHN腎癌干細(xì)胞的凋亡,各組凋亡率分別為:空白對(duì)照組0.55±0.075%;低濃度組:3.48±0.505%;中濃度組:9.33±0.612%;高濃度組:18.36±1.511%,各組間比較P值均0.05,差異有統(tǒng)計(jì)學(xué)意義。3.三種濃度的“益氣解毒方”藥物血清均不同程度的上調(diào)了ACHN腎癌干細(xì)胞的P53蛋白的表達(dá),分別為空白對(duì)照組:0.78±0.25%;低濃度組:7.24±0.80%;中濃度組:11.72±1.19%;高濃度組:17.68±1.06%;各組間比較P值均0.05,差異有統(tǒng)計(jì)學(xué)意義。結(jié)論:1.“益氣解毒方”藥物血清可以抑制ACHN腎癌干細(xì)胞增殖。2.“益氣解毒方”藥物血清可以誘導(dǎo)ACHN腎癌干細(xì)胞凋亡。3.“益氣解毒方”藥物血清可以提高ACHN腎癌干細(xì)胞P53蛋白的表達(dá)。
[Abstract]:Objective: to investigate the effect of Yiqi jiedu recipe on proliferation and apoptosis of ACHN renal cell carcinoma stem cells. Methods: high, medium and low concentrations of "Yiqi jiedu recipe" were used to intervene ACHN renal cell carcinoma stem cells in vitro. The effect of "Yiqi jiedu recipe" on the proliferation of ACHN renal cell carcinoma stem cells was detected by dimethyl azo blue (MTT) assay. Apoptosis of ACHN renal cell carcinoma stem cells was detected by flow cytometry under the intervention of "Yiqi jiedu recipe" serum. The expression of p53 protein in ACHN renal cell carcinoma stem cells was detected by flow cytometry. Result: 1.
