芪桂顆粒對甲狀腺功能減退大鼠IL-1及IFN-γ的影響

【摘要】:目的:本實驗通過觀察自擬方芪桂顆粒對甲減大鼠血清IL-1及IFN-γ表達水平影響,以觀察芪桂顆粒治療甲減的治療效果,探討其對下丘腦-垂體-甲狀腺軸及相關(guān)細胞因子的調(diào)控作用,并闡明其作用機理。方法:選取健康清潔級Wistar大鼠50只,隨機分為空白組、模型組、西藥組、中藥低劑量組和中藥高劑量組5組,每組10只?瞻捉M給予生理鹽水灌胃,其余各組均給予0.1%丙硫氧嘧啶(Propylthiouracil,PTU)生理鹽水溶液灌胃,造甲減大鼠模型。15天后采血檢測甲狀腺功能及IL-1、IFN-γ含量,證實模型建立成功,后給予相應(yīng)的西藥、芪桂顆粒連續(xù)治療8周。8周后統(tǒng)一采血,采用電化學(xué)發(fā)光法檢測FT3、FT4、TSH、TG-Ab、TPO-Ab。采用ELISA試劑盒檢測IL-1、IFN-γ。結(jié)果:造模后的甲減大鼠在給藥治療后,其甲減癥狀均有不同程度的改善。給藥8周之后,中藥各劑量組FT3、FT4值均高于模型組,有顯著性差異(P0.05);中藥各劑量組TSH值均低于模型組,有顯著性差異(P0.05);中藥高劑量組的FT3、FT4、TSH值與中藥低劑量組比較,有極顯著性差異(P0.01);中藥高劑量組的FT3、FT4、TSH值與西藥組比較,無統(tǒng)計學(xué)差異(P0.05)。中藥低、高劑量組IL-1、IFN-γ均明顯低于模型組,有顯著性差異(P0.05);中藥低劑量組與西藥組IL-1、IFN-γ值比較,無統(tǒng)計學(xué)差異(P0.05);中藥高劑量組與空白組IL-1、IFN-γ值比較,無統(tǒng)計學(xué)差異(P0.05);中藥高劑量組與西藥組IL-1、IFN-γ值比較,有極顯著性差異(P0.01)。給藥后,各組大鼠血清TG-Ab、TPO-Ab改善不顯著,無統(tǒng)計學(xué)差異(P0.05)。結(jié)論:芪桂顆?梢燥@著改善甲減大鼠一般狀態(tài),升高FT3、FT4水平,降低TSH水平。芪桂顆粒高劑量組可以顯著降低IL-1、IFN-γ水平,改善大鼠甲減癥狀。
[Abstract]:Objective: to observe the effect of Qigi granule on the expression of IL-1 and IFN- 緯 in serum of hypothyroidism rats. To investigate the regulation of hypothalamus-pituitary-thyroid axis and related cytokines, and to elucidate its mechanism. Methods: 50 healthy and clean grade Wistar rats were randomly divided into blank group, model group, western medicine group, low dose Chinese medicine group and high dose Chinese medicine group with 10 rats in each group. The rats in the blank group were given normal saline and the other groups were given 0.1% Propylthiouracil,PTU saline solution to make the model of hypothyroidism. After 15 days, the thyroid function and the content of IL-1,IFN- 緯 were measured. It was proved that the model was established successfully, and then the corresponding western medicine was given. Qigui granule was treated continuously for 8 weeks. After 8 weeks, blood was collected uniformly, and FT3,FT4,TSH,TG-Ab,TPO-Ab. was detected by electrochemiluminescence method. ELISA kit was used to detect IL-1,IFN- 緯. Results: the hypothyroidism symptoms of hypothyroidism rats were improved in varying degrees. After 8 weeks of administration, the FT3,FT4 value of each dose group was higher than that of the model group (P0.05), and the TSH value of each dose group was lower than that of the model group (P0.05). The FT3,FT4,TSH value of the high dose group was significantly higher than that of the low dose group (P0.01), while the FT3,FT4,TSH value of the high dose group was not significantly different from that of the western medicine group (P0.05). IL-1,IFN- 緯 in low and high dose group was significantly lower than that in model group (P0.05), but there was no significant difference in IL-1,IFN- 緯 between low dose group and western medicine group (P0.05). There was no significant difference in IL-1,IFN- 緯 between the high dose group and the blank group (P0.05); the IL-1,IFN- 緯 value in the high dose group was significantly higher than that in the western medicine group (P0.01). After administration of the drug, the improvement of serum TG-Ab,TPO-Ab in each group was not significant, there was no statistical difference (P0.05). Conclusion: Qigui granule can significantly improve the general state of hypothyroidism rats, increase the level of FT3,FT4 and decrease the level of TSH. Qigui granule high dose group can significantly reduce the level of IL-1,IFN- 緯, improve the symptoms of hypothyroidism in rats.
