【摘要】：目的:觀察電針"曲池""足三里"穴對高血壓前期Dahl鹽敏感(DS)大鼠腎臟NO/c GMP/PKG信號通路的影響及組織形態(tài)的改變,探討電針"曲池""足三里"對高血壓前期腎臟損害保護(hù)作用機(jī)制。方法:鹽抵抗大鼠(DR)10只作為正常組;DS大鼠30只,隨機(jī)分為模型組、針刺組和非穴位組,每組10只。采用高鹽飼料喂養(yǎng)的方法誘發(fā)高血壓前期模型。電針組電針"曲池""足三里"穴,1次/d,6次/周,共5周。非穴位組取雙側(cè)髂嵴上10~15 mm、后正中線旁開20mm區(qū)段內(nèi)一個(gè)固定對照點(diǎn),治療方法同電針組。測量大鼠治療期間血壓變化情況以及治療后血清血清尿素氮(UREA)、血肌酐(CREA)、血尿酸(UA)含量;ELISA法檢測NO含量;熒光定量PCR檢測大鼠腎組織溶解型鳥苷酸環(huán)化酶(s GC)、蛋白激酶G(PKG)的mRNA表達(dá);HE染色檢測大鼠腎組織病理形態(tài)變化。結(jié)果:治療期間,電針組大鼠血壓逐步下降。與正常組相比,模型組血清UREA、CREA、UA含量顯著升高(P0.05),NO含量顯著降低(P0.05),s GC、PKG mRNA表達(dá)顯著降低(P0.05);與模型組相比,電針組血清UREA、CREA、UA含量顯著降低(P0.05),NO含量顯著升高(P0.05),s GC、PKG mRNA表達(dá)顯著升高(P0.05)。結(jié)論:電針能有效降低高血壓前期DS大鼠血壓,提高NO含量、c GMP、PKG基因表達(dá),改善組織小管擴(kuò)張、間質(zhì)炎癥細(xì)胞浸潤等,這可能是電針通過NO/c GMP/PKG信號通路發(fā)揮對高血壓前期DS大鼠腎臟保護(hù)作用機(jī)制之一。
[Abstract]:Objective: to observe the effect of electroacupuncture at "qu Chi" and "Zusanli" on the renal NO/c GMP/PKG signaling pathway and the changes of tissue morphology in pre-hypertensive Dahl salt-sensitive (DS) rats. To explore the protective mechanism of electroacupuncture (EA) on renal damage in prehypertensive stage of Zusanli (Zusanli). Methods: ten (DR) rats with salt resistance and 30 DS rats were randomly divided into three groups: model group, acupuncture group and non-acupoint group with 10 rats in each group. The prehypertension model was induced by high salt feed. In the electroacupuncture group, the acupoints of Zusanli, once a day, 6 times a week for 5 weeks. After 1015 mm, of bilateral iliac crest, a fixed control point in 20mm segment was opened in the non-acupoint group. The treatment method was the same as that in the electroacupuncture group. Serum urea nitrogen (UREA), serum creatinine (CREA), serum uric acid (UA), serum urea nitrogen (UREA), serum creatinine (CREA), serum uric acid (UA) were measured in rats during treatment, and NO content was detected by ELISA. Fluorescence quantitative PCR was used to detect the mRNA expression of (s GC), protein kinase G (PKG) in rat renal tissue, and HE staining was used to detect the histopathological changes of renal tissue. Results: during the treatment period, the blood pressure of the rats in the electroacupuncture group decreased gradually. Compared with the normal group, the serum UREA,CREA,UA content in the model group was significantly increased (P0.05), NO content significantly decreased (P0.05), s GC,PKG mRNA expression significantly decreased (P0.05); Compared with the model group, the serum UREA,CREA,UA content in the electroacupuncture group was significantly lower (P0.05), NO content was significantly higher (P0.05), s GC,PKG mRNA expression was significantly higher (P0.05). Conclusion: electroacupuncture can effectively reduce blood pressure, increase NO content, c GMP,PKG gene expression, improve tubule dilatation and interstitial inflammatory cell infiltration in prehypertensive DS rats. This may be one of the mechanisms of renal protection by electroacupuncture through NO/c GMP/PKG signaling pathway in prehypertensive DS rats.
