艾灸肺俞、膏肓俞治療肺纖維化大鼠對E-cad、α-SMA、vimentin的影響

【摘要】：目的:動態(tài)觀察艾灸肺俞、膏肓俞治療肺纖維化大鼠E-鈣黏蛋白(E-cad)、α-平滑肌蛋白(α-SMA)、波形蛋白(vimentin)幾種上皮-間質(zhì)轉(zhuǎn)分化(EMT)標(biāo)志物的表達。方法:選用SD雄性大鼠44只,隨機分為4組:正常對照組、模型組、艾灸治療組、潑尼松治療組。除正常對照組外,其余三組均采用博來霉素灌入氣管復(fù)制肺纖維化模型,治療組從造模后第1天開始分別艾灸及潑尼松灌胃治療,在造模后第11天、第23天、第35天取材。采用HE染色、masson染色觀察肺組織病理變化和采用免疫組化法檢測E-cad、α-SMA、vimentin的表達。結(jié)果:艾灸組、潑尼松組與同時段模型組相比,肺組織病理程度明顯降低,上皮細(xì)胞標(biāo)記物E-cad表達升高,間質(zhì)細(xì)胞標(biāo)記物α-SMA、vimentin表達降低。結(jié)論:艾灸治療能升高E-cad水平,降低α-SMA、vimentin水平,降低肺泡炎程度及延緩肺纖維化進程。
[Abstract]:Aim: to observe the expression of E-cadherin (E-cad) and 偽 -smooth muscle protein (偽 SMA), vimentin (vimentin) in the treatment of pulmonary fibrosis rats with moxibustion and ointment. Methods: 44 male SD rats were randomly divided into 4 groups: normal control group, model group, moxibustion group and prednisone group. Except the normal control group, the other three groups were treated with bleomycin into the trachea to make pulmonary fibrosis model. The treatment group was treated with moxibustion and prednisone respectively on the 1st day after the model was made. The materials were obtained on the 11th, 23rd and 35th day after the model was made. HE staining and masson staining were used to observe the pathological changes of lung tissue and the expression of E-cad- 偽-SMA,vimentin was detected by immunohistochemical method. Results: in moxibustion group and prednisone group, compared with the model group, the pathological degree of lung tissue was significantly decreased, the expression of E-cad and 偽-SMA,vimentin, the marker of epithelial cells were increased, and the expression of 偽-SMA,vimentin in interstitial cells was decreased significantly in the moxibustion group and prednisone group. Conclusion: moxibustion treatment can increase the level of E-cad, decrease the level of 偽-SMA,vimentin, decrease the degree of alveolitis and delay the progress of pulmonary fibrosis.
