針刺對DMBA誘導的乳腺癌大鼠ER、PR表達的影響
[Abstract]:Aim: to observe the effect of acupuncture on the expression of estrogen receptor (ER),) progesterone receptor (PR) in breast tissues of rats with breast cancer induced by dimethyl benzene anthracene (DMBA), and to explore the mechanism of acupuncture in preventing and treating breast cancer. Methods: 120 SD female rats aged 6 to 8 weeks were randomly divided into model group (n = 90) and blank group (n = 30). In the model group, DMBA was used to make the model, and the blank group was given the same dose of sesame oil. After 15 weeks, the rats in the model group were randomly divided into treatment group and control group. The treatment group was treated with acupuncture at Zusanli, Sanyinjiao and Tanzhong three times a week for 20 mins for 12 weeks. Control group and blank group did not receive any treatment, only at the same time. At the end of acupuncture (27 weeks), the serum tumor markers were detected by electrochemiluminescence immunoassay (ECI). The number of tumor masses was counted and the shape was recorded, and the tissue of the tumor was taken. The height, maximum diameter and vertical diameter of the mass were measured with Vernier caliper with precision of 1mm. The histopathological changes, positive area and average optical density of ER,PR were observed under Olympus optical microscope. Results: the mean number and volume of breast tumors in the treatment group were significantly different from those in the control group and the blank group (P 0.05). The mean number and volume of breast tumors in the control group were significantly different from those in the blank group (P0.01). The concentrations of tumor markers (CA724,CA125,CA199,AEP,CA15-3,CEA and CA50) in the treatment group and the control group were significantly different from those in the blank group (P 0.05). The concentration of CA15-3 in the treatment group was significantly higher than that in the control group (P0.01). The positive area and average optical density of ER,PR in the treatment group were significantly higher than those in the control group and the blank group (P0.01). The positive area and average optical density of ER,PR in the control group were significantly different from those in the control group (P0.01). Conclusion: acupuncture can reduce the occurrence of breast tumor (including tumor number and tumor volume) induced by DMBA in rats. The mechanism of acupuncture may be related to decreasing the concentration of tumor markers such as CA724,CA125,CA199,CA15-3,AEP,CEA,CA50. Especially, decreasing the concentration of CA15-3 and decreasing the positive area of ER,PR were related to the average optical density.
