【摘要】：目的:觀(guān)察針刺療法對(duì)二甲基苯蒽(DMBA)誘導(dǎo)的乳腺癌大鼠乳腺組織中雌激素受體(ER)、孕激素受體(PR)表達(dá)的影響,探索針刺防治乳腺癌的作用機(jī)制。方法:將120只6~8周齡的SD雌性大鼠隨機(jī)分為模型組90只和空白組30只。模型組采用DMBA進(jìn)行灌胃造模,空白組給予等劑量的芝麻油灌胃。造模15周后,將模型組大鼠隨機(jī)分為治療組和對(duì)照組。治療組采用針刺足三里、三陰交、膻中治療,每周3次,每次20min,共治療12周。對(duì)照組和空白組不采用任何治療,僅在相同時(shí)間給予抓放。針刺治療結(jié)束后(第27周),采用電化學(xué)發(fā)光免疫方法,取腹腔靜脈血做血清腫瘤標(biāo)志物檢測(cè);計(jì)數(shù)腫瘤包塊數(shù)量并記錄形態(tài),取包塊組織,用精度為1mm的游標(biāo)卡尺測(cè)量包塊的高度、最大直徑和相垂直的直徑;采用Olympus光學(xué)顯微鏡下觀(guān)察腫瘤組織病理學(xué)變化及ER、PR的陽(yáng)性面積及平均光密度。結(jié)果:治療組治療后乳腺腫瘤平均數(shù)目和平均體積與對(duì)照組和空白組比較,差異均具有統(tǒng)計(jì)學(xué)意義(P0.01,P0.05);對(duì)照組治療后乳腺腫瘤平均數(shù)目和平均體積與空白組比較,差異均具有統(tǒng)計(jì)學(xué)意義(P0.01);治療組和對(duì)照組治療后各項(xiàng)腫瘤標(biāo)志物(CA724、CA125、CA199、AEP、CA15-3、CEA及CA50)濃度與空白組比較,差異均具有統(tǒng)計(jì)學(xué)意義(P0.01,P0.05);治療組治療后CA15-3濃度與對(duì)照組比較,差異具有統(tǒng)計(jì)學(xué)意義(P0.01);治療組治療后ER、PR陽(yáng)性面積和平均光密度與對(duì)照組和空白組比較,差異均具有統(tǒng)計(jì)學(xué)意義(P0.01);對(duì)照組治療后ER、PR陽(yáng)性面積和平均光密度與空白組比較,差異均具有統(tǒng)計(jì)學(xué)意義(P0.01)。結(jié)論:針刺能夠減少DMBA誘導(dǎo)的大鼠乳腺腫瘤的發(fā)生(包括腫瘤數(shù)目及腫瘤體積的大小),其作用機(jī)制可能與降低CA724、CA125、CA199、CA15-3、AEP、CEA、CA50等腫瘤標(biāo)志物的濃度,尤其是降低CA15-3的濃度,減少ER、PR的陽(yáng)性面積和平均光密度相關(guān)。
[Abstract]:Aim: to observe the effect of acupuncture on the expression of estrogen receptor (ER),) progesterone receptor (PR) in breast tissues of rats with breast cancer induced by dimethyl benzene anthracene (DMBA), and to explore the mechanism of acupuncture in preventing and treating breast cancer. Methods: 120 SD female rats aged 6 to 8 weeks were randomly divided into model group (n = 90) and blank group (n = 30). In the model group, DMBA was used to make the model, and the blank group was given the same dose of sesame oil. After 15 weeks, the rats in the model group were randomly divided into treatment group and control group. The treatment group was treated with acupuncture at Zusanli, Sanyinjiao and Tanzhong three times a week for 20 mins for 12 weeks. Control group and blank group did not receive any treatment, only at the same time. At the end of acupuncture (27 weeks), the serum tumor markers were detected by electrochemiluminescence immunoassay (ECI). The number of tumor masses was counted and the shape was recorded, and the tissue of the tumor was taken. The height, maximum diameter and vertical diameter of the mass were measured with Vernier caliper with precision of 1mm. The histopathological changes, positive area and average optical density of ER,PR were observed under Olympus optical microscope. Results: the mean number and volume of breast tumors in the treatment group were significantly different from those in the control group and the blank group (P 0.05). The mean number and volume of breast tumors in the control group were significantly different from those in the blank group (P0.01). The concentrations of tumor markers (CA724,CA125,CA199,AEP,CA15-3,CEA and CA50) in the treatment group and the control group were significantly different from those in the blank group (P 0.05). The concentration of CA15-3 in the treatment group was significantly higher than that in the control group (P0.01). The positive area and average optical density of ER,PR in the treatment group were significantly higher than those in the control group and the blank group (P0.01). The positive area and average optical density of ER,PR in the control group were significantly different from those in the control group (P0.01). Conclusion: acupuncture can reduce the occurrence of breast tumor (including tumor number and tumor volume) induced by DMBA in rats. The mechanism of acupuncture may be related to decreasing the concentration of tumor markers such as CA724,CA125,CA199,CA15-3,AEP,CEA,CA50. Especially, decreasing the concentration of CA15-3 and decreasing the positive area of ER,PR were related to the average optical density.
