凋瘤丸對乳腺癌細(xì)胞MCF-7凋亡基因Caspase-3的影響

【摘要】:目的:本文就探討凋瘤丸對體外培養(yǎng)人乳腺癌細(xì)胞株MCF-7的誘導(dǎo)凋亡作用及凋亡相關(guān)蛋白Caspase-3表達(dá)的影響的論述。希望能為今后研究中藥抗癌機(jī)制提供依據(jù)和思路,為臨床上治療乳腺癌提供一定的分子生物學(xué)依據(jù),以及為該藥在臨床的應(yīng)用提供相應(yīng)的理論基礎(chǔ)。方法:用含10%胎牛血清,1%鏈霉素、青霉素的MEM養(yǎng)液,于37℃,5%CO2孵箱中培養(yǎng)人乳腺癌MCF-7細(xì)胞株;1光鏡形態(tài)學(xué)觀察:加入凋瘤丸提取液培養(yǎng)48h后,取出細(xì)胞片,常規(guī)HE染色,運用光學(xué)顯微鏡觀察并記錄;2 MTT實驗:將MCF-7細(xì)胞按1×105個/孔接種于96孔板,設(shè)置3個復(fù)孔,隨機(jī)分為:凋瘤丸懸液高、中、低濃度藥物組,陰性對照組,三苯氧胺對照組。進(jìn)行MTT試驗,計算出細(xì)胞增殖抑制率,篩選出最合適的藥物劑量;3 Hoechst33258檢測試驗:取指數(shù)生長期的細(xì)胞,運Hoechst33258染色法熒光倒置顯微鏡觀察調(diào)瘤丸藥液作用后人乳腺癌細(xì)胞株MCF-7的早期凋亡情況。4 RT-PCR檢測將凋瘤丸懸液高、中、低濃度藥物組,陰性對照組(不做任何治療),三苯氧胺對照組三組分別培養(yǎng)作用48小時后,經(jīng)胰酶消化收集細(xì)胞,然后提取細(xì)胞MCF-7的RNA。檢測在不同分組中caspase-3基因表達(dá)量的變化。結(jié)果顯示:在光學(xué)顯微鏡下,凋瘤丸提取液作用過細(xì)胞體積變小、形態(tài)變圓,核被染為深藍(lán)色,胞漿呈淺紅色,核呈破碎狀,染色質(zhì)顏色凝集加深,凋亡小體形成等;Hoechst染色檢測顯示;低濃度凋瘤丸組僅見有核凹陷以及小塊缺損,但結(jié)構(gòu)完整;隨著濃度的增加,凋亡小體(呈現(xiàn)胞質(zhì)胞核的皺縮,染色質(zhì)邊集濃染,細(xì)胞核被濃染而發(fā)出亮藍(lán)色強(qiáng)烈熒光,呈現(xiàn)出“新月狀”、帶狀甚至破碎斑塊狀,又稱凋亡小體)而增多,陽性對照組與實驗組有相似的作用效果;MTT檢測法提示:陰性對照組細(xì)胞存活率極高,增殖抑制率接近0;高中低凋瘤丸藥物組以及三苯氧胺組均對MCF-7細(xì)胞的增殖均起到不同程度的抑制;而且,這種抑制程度隨著藥物濃度的遞增而遞增。最大劑量的凋瘤丸組與三苯氧胺組的效果接近。RT-PCR檢測結(jié)果顯示,與陰性對照組相比,各藥物處理后的MCF-7細(xì)胞中caspase-3均表現(xiàn)為上調(diào)表達(dá),其中凋瘤丸懸液高濃度組和三苯氧胺對照組與陰性對照組相比,具有統(tǒng)計學(xué)意義,(P0.05)。而凋瘤丸懸液中濃度組和凋瘤丸懸液低濃度組無統(tǒng)計學(xué)意義。凋瘤丸能抑制MCF-7細(xì)胞株的增殖,且呈量效的依賴性。最大濃度的結(jié)果接近三苯氧胺組無統(tǒng)計學(xué)差異(p0.05),與陰性對照組有統(tǒng)計學(xué)差異(P0.05)。結(jié)論:1.凋瘤丸對乳腺癌細(xì)胞有誘導(dǎo)凋亡作用,且在一定的范圍內(nèi),這種抑瘤作用呈量效關(guān)系。2.凋瘤丸藥物組可以影響caspase-3的表達(dá),使其上調(diào),且通過激活Caspase-3的表達(dá)來誘導(dǎo)乳腺癌細(xì)胞凋亡。
