針刺對支氣管哮喘大鼠肺組織CollagenⅠ、Fibronection、VEGF表達的影響
[Abstract]:Objective to observe the effect of acupuncture on the expression of Col in lung tissue of asthmatic rats, in order to clarify the relationship between Col and asthma and the regulatory effect of acupuncture on it. Methods 90 rats were randomly divided into 3 groups: blank group, model group and acupuncture group. Asthma models were induced by intraperitoneal injection of OVA and Al (OH) _ 3 and then inhaled by OVA solution. The blank group inhaled normal saline, and the acupuncture group inhaled OVA solution after acupuncture. According to the difference of total intervention time, 10 rats were selected from each group according to random number table method after 4 weeks of atomization, and the remaining rats were killed after 6 weeks of atomization. The rats in the blank group were recorded as Acarinic at 6 weeks, the rats in the model group in the atomized group at 2 and 4 weeks were recorded as DGV EfU, and the rats in the acupuncture group were recorded as GG HG I at the 6th week of acupuncture. The pathological changes of lung tissue were observed by HE staining and Masson staining, and the expression of Col IfNNtr in lung tissue of each group was observed by immunohistochemical method and ELISA method. Results 1 the rats in the model group had symptoms such as agitation, rapid breathing, cyanosis and incontinence when they were stimulated by agonist atomization, and the rats in the model group had symptoms such as restlessness, rapid breathing, cyanosis of the lips, and incontinence. The above symptoms were mild in acupuncture group and blank group. (2) HE staining in lung tissue of rats in each group showed severe airway remodeling and pathological changes of airway inflammation in model group, and with the increase of atomization cycle, the lung tissue of model group showed severe pathological changes of airway remodeling and airway inflammation. The pathological changes of lung tissue in group F were increased. There were no obvious pathological changes in lung tissue in acupuncture group and blank group, and there was no obvious change in lung tissue structure with the increase of atomization cycle. 3 Masson staining of lung tissue in each group showed that WA/Pi,WAm/Pi,WAc/Pi, in lung tissue of model group was not changed. Compared with the blank group, the average value of WAi/Pi increased (P0.05). With the increase of atomization cycle, the average value of WA/Pi,WAm/Pi,WAi/Pi in DIAE E F group showed an increasing trend, and WAc/Pi did not increase with the increase of atomization cycle. There was no significant difference in WA/Pi,WAm/Pi,WAc/Pi,WAi/Pi between the acupuncture group and the blank group (P0.05). 4 the lung tissue of each group was detected by immunohistochemical method and ELISA method. The positive expression of VEGF was significantly higher than that of the blank group (P0.05), and the expression of VEGF in Col Igna FNN was decreased with the prolongation of the atomization cycle, but still higher than that in the blank group (P0.05), the difference was statistically significant. The expression of Col in lung tissue in acupuncture group was lower than that in model group (P0.05). Conclusion (1) Acupuncture can significantly improve the symptoms of asthmatic rats induced by ovalbumin atomization, such as dyspnea, purplish lip, agitation, etc. The degree of airway remodeling in group F was gradually aggravated, while acupuncture could significantly inhibit the development of airway remodeling in asthmatic rats. 3 the expression of Col in lung tissue of asthmatic rats was increased. The expression of Col in lung tissue of asthmatic rats was decreased with the increase of the times of atomization stimulation. 4 Acupuncture could inhibit the expression of Col in lung tissue of asthmatic rats.
