針刺對(duì)支氣管哮喘大鼠肺組織CollagenⅠ、Fibronection、VEGF表達(dá)的影響

【摘要】:目的通過(guò)觀(guān)察針刺對(duì)支氣管哮喘大鼠肺組織Col I、FN、VEGF表達(dá)的影響,以期明確Col I、FN、VEGF與哮喘的關(guān)系以及針刺對(duì)其的調(diào)控作用。方法將90只大鼠按照隨機(jī)數(shù)字表法分成3組,每組30只,分別為空白組、模型組、針刺組。采用腹腔注射OVA和Al(OH)3混合液致敏后霧化吸入OVA溶液激發(fā)法制備哮喘模型?瞻捉M霧化吸入生理鹽水,針刺組于針刺后霧化吸入OVA溶液。根據(jù)總干預(yù)時(shí)間的差異,分別于霧化2、4周后,按照隨機(jī)數(shù)字表法從每組大鼠中選出10只處死,于霧化6周后處死剩余大鼠?瞻捉M霧化2、4、6周大鼠分別記為A、B、C,模型組霧化2、4、6周大鼠分別記為D、E、F,針刺組針刺2、4、6周大鼠分別記為G、H、I。通過(guò)HE染色、Masson染色觀(guān)察各組大鼠肺組織的病理改變情況,通過(guò)免疫組化法、ELISA法觀(guān)察各組大鼠肺組織Col I、FN、VEGF的表達(dá)情況。結(jié)果1模型組大鼠在接受激動(dòng)劑霧化激發(fā)時(shí),出現(xiàn)煩躁不安、呼吸加快加深、口唇紫紺、二便失禁等癥狀,針刺組和空白組大鼠在霧化時(shí)上述癥狀較輕。2各組大鼠肺組織HE染色顯示,模型組大鼠肺組織出現(xiàn)嚴(yán)重的氣道重塑及氣道炎癥病理改變,且隨著霧化周期的增加D、E、F組肺組織病理改變程度增加。針刺組及空白組大鼠肺組織無(wú)明顯病理改變,且隨著霧化周期的增加肺組織結(jié)構(gòu)無(wú)明顯變化。3各組大鼠肺組織Masson染色顯示,模型組大鼠肺組織WA/Pi、WAm/Pi、WAc/Pi、WAi/Pi平均值均較空白組增大(P0.05),隨著霧化周期的增長(zhǎng),D、E、F組WA/Pi、WAm/Pi、WAi/Pi的均值呈增長(zhǎng)趨勢(shì),WAc/Pi無(wú)隨霧化周期增長(zhǎng)而增厚的現(xiàn)象。針刺組與空白組大鼠肺組織WA/Pi、WAm/Pi、WAc/Pi、WAi/Pi平均值無(wú)顯著差異(P0.05)。4各組大鼠肺組織免疫組化法、ELISA法檢測(cè)顯示,模型組大鼠肺組織Col I、FN、VEGF的陽(yáng)性表達(dá)明顯高于空白組(P0.05),且隨著霧化周期的延長(zhǎng)D、E、F各組Col I、FN、VEGF的表達(dá)呈下降趨勢(shì),但仍較空白組高(P0.05),差異有統(tǒng)計(jì)學(xué)意義。針刺組大鼠肺組織Col I、FN、VEGF的表達(dá)低于模型組(P0.05),差異有統(tǒng)計(jì)學(xué)意義。結(jié)論1針刺可以顯著改善卵蛋白霧化激發(fā)引起的哮喘大鼠呼吸困難、口唇青紫、煩躁不安等癥狀。2隨著霧化激發(fā)周期的增加,模型組D、E、F小組大鼠肺組織氣道重塑程度逐漸加重,而針刺可以明顯抑制哮喘大鼠氣道重塑的發(fā)生發(fā)展。3哮喘模型大鼠肺組織Col I、FN、VEGF的表達(dá)增多,且隨著霧化激發(fā)次數(shù)的增多哮喘大鼠肺組織Col I、FN、VEGF的表達(dá)水平呈下降趨勢(shì)。4針刺可以抑制哮喘大鼠肺組織Col I、FN、VEGF的表達(dá)。
[Abstract]:Objective to observe the effect of acupuncture on the expression of Col in lung tissue of asthmatic rats, in order to clarify the relationship between Col and asthma and the regulatory effect of acupuncture on it. Methods 90 rats were randomly divided into 3 groups: blank group, model group and acupuncture group. Asthma models were induced by intraperitoneal injection of OVA and Al (OH) _ 3 and then inhaled by OVA solution. The blank group inhaled normal saline, and the acupuncture group inhaled OVA solution after acupuncture. According to the difference of total intervention time, 10 rats were selected from each group according to random number table method after 4 weeks of atomization, and the remaining rats were killed after 6 weeks of atomization. The rats in the blank group were recorded as Acarinic at 6 weeks, the rats in the model group in the atomized group at 2 and 4 weeks were recorded as DGV EfU, and the rats in the acupuncture group were recorded as GG HG I at the 6th week of acupuncture. The pathological changes of lung tissue were observed by HE staining and Masson staining, and the expression of Col IfNNtr in lung tissue of each group was observed by immunohistochemical method and ELISA method. Results 1 the rats in the model group had symptoms such as agitation, rapid breathing, cyanosis and incontinence when they were stimulated by agonist atomization, and the rats in the model group had symptoms such as restlessness, rapid breathing, cyanosis of the lips, and incontinence. The above symptoms were mild in acupuncture group and blank group. (2) HE staining in lung tissue of rats in each group showed severe airway remodeling and pathological changes of airway inflammation in model group, and with the increase of atomization cycle, the lung tissue of model group showed severe pathological changes of airway remodeling and airway inflammation. The pathological changes of lung tissue in group F were increased. There were no obvious pathological changes in lung tissue in acupuncture group and blank group, and there was no obvious change in lung tissue structure with the increase of atomization cycle. 3 Masson staining of lung tissue in each group showed that WA/Pi,WAm/Pi,WAc/Pi, in lung tissue of model group was not changed. Compared with the blank group, the average value of WAi/Pi increased (P0.05). With the increase of atomization cycle, the average value of WA/Pi,WAm/Pi,WAi/Pi in DIAE E F group showed an increasing trend, and WAc/Pi did not increase with the increase of atomization cycle. There was no significant difference in WA/Pi,WAm/Pi,WAc/Pi,WAi/Pi between the acupuncture group and the blank group (P0.05). 4 the lung tissue of each group was detected by immunohistochemical method and ELISA method. The positive expression of VEGF was significantly higher than that of the blank group (P0.05), and the expression of VEGF in Col Igna FNN was decreased with the prolongation of the atomization cycle, but still higher than that in the blank group (P0.05), the difference was statistically significant. The expression of Col in lung tissue in acupuncture group was lower than that in model group (P0.05). Conclusion (1) Acupuncture can significantly improve the symptoms of asthmatic rats induced by ovalbumin atomization, such as dyspnea, purplish lip, agitation, etc. The degree of airway remodeling in group F was gradually aggravated, while acupuncture could significantly inhibit the development of airway remodeling in asthmatic rats. 3 the expression of Col in lung tissue of asthmatic rats was increased. The expression of Col in lung tissue of asthmatic rats was decreased with the increase of the times of atomization stimulation. 4 Acupuncture could inhibit the expression of Col in lung tissue of asthmatic rats.
