三伏灸誘導(dǎo)胸腔體表不同灸溫對哮喘型新西蘭兔肺組織MMP-9表達(dá)的影響

【摘要】:目的:對比觀察三伏灸不同灸溫對哮喘型新西蘭兔肺組織MMP-9蛋白及mRNA表達(dá)的影響差異,并探討胸腔體表溫度的變化與隔姜灸量的相關(guān)性,及其與氣至病所的關(guān)系。方法:選擇雄性新西蘭兔30只,體質(zhì)量(2.5±0.3)kg。采用隨機(jī)數(shù)字表隨機(jī)分為空白組,模型組,實(shí)驗(yàn)組,對照組,地塞米松組,每組6只?瞻捉M不造模,其余各組均按照哮喘氣道重塑模型制作方法進(jìn)行造模。模型組僅造模,不施予治療。實(shí)驗(yàn)組及對照組均給予隔姜灸聯(lián)合中藥敷貼干預(yù),穴位均選擇大椎及雙側(cè)肺俞,并采用紅外熱像儀監(jiān)測兩組哮喘型新西蘭兔胸腔體表溫度變化,兩組均干預(yù)15次,隔日1次。實(shí)驗(yàn)組予隔姜灸至胸腔體表溫度較治療前升高1℃,對照組予隔姜灸3壯干預(yù)。地塞米松組給予0.5mg/kg地塞米松霧化吸入,干預(yù)15次,隔日1次。實(shí)驗(yàn)結(jié)束后,取新西蘭兔右肺組織,采用免疫組織化學(xué)染色方法,檢測右肺組織MMP-9蛋白表達(dá)。采用熒光定量PCR法檢測左肺組織MMP-9 mRNA表達(dá)。觀察5組新西蘭兔MMP-9的改變情況,同時(shí)分析胸腔體表溫度的變化與隔姜灸量的相關(guān)性,及其與氣至病所的關(guān)系。結(jié)果:1.模型組、實(shí)驗(yàn)組、對照組、地塞米松組的MMP-9蛋白及mRNA表達(dá)明顯高于空白組,差異均具有統(tǒng)計(jì)學(xué)意義(P0.05);實(shí)驗(yàn)組、對照組、地塞米松組的MMP-9蛋白及mRNA表達(dá)明顯低于模型組,差異均具有統(tǒng)計(jì)學(xué)意義(P0.05);實(shí)驗(yàn)組的MMP-9蛋白及mRNA表達(dá)明顯低于對照組,差異均具有統(tǒng)計(jì)學(xué)意義(P0.05);實(shí)驗(yàn)組與地塞米松組的MMP-9蛋白及mRNA表達(dá)無明顯差異,差異無統(tǒng)計(jì)學(xué)意義(P0.05)。2.實(shí)驗(yàn)組灸后的胸腔體表溫度明顯高于灸前的體表溫度,差異具有統(tǒng)計(jì)學(xué)意義(P0.05);對照組灸后的胸腔體表溫度與灸前無明顯差異,差異無統(tǒng)計(jì)學(xué)意義(P0.05);實(shí)驗(yàn)組灸后的胸腔體表溫度明顯高于對照組灸后的胸腔體表溫度,差異具有統(tǒng)計(jì)學(xué)意義(P0.05);實(shí)驗(yàn)組的胸腔體表溫度隨著隔姜灸壯數(shù)的增加而升高,spearman相關(guān)系數(shù)r=0.433,具有顯著性差異(P0.01)。結(jié)論:隔姜灸達(dá)到9壯及以上可能通過升高哮喘型新西蘭兔胸腔體表溫度而誘導(dǎo)出氣至病所,從而下調(diào)肺組織MMP=9蛋白及mRNA表達(dá),抑制MMP-9的合成,改善哮喘型新西蘭兔氣道重塑,從而有效地緩解哮喘癥狀。同時(shí),胸腔體表溫度與隔姜灸的壯數(shù)呈正相關(guān),表明溫度的提升可能為隔姜灸時(shí)達(dá)到氣至病所的相關(guān)指標(biāo)之一。
[Abstract]:Objective: to observe the effect of different moxibustion temperature on the expression of MMP-9 protein and mRNA in lung tissue of asthmatic New Zealand rabbits, and to explore the relationship between the surface temperature of thoracic cavity and the quantity of ginger separated moxibustion. Methods: 30 male New Zealand rabbits with body weight of (2.5 鹵0.3) kg. were selected. Six rats were randomly divided into blank group, model group, experimental group, control group and dexamethasone group. No model was made in the blank group, and the other groups were made according to the airway remodeling model of asthma. In the model group, only the model was made and not treated. The experimental group and the control group were treated with ginger separated moxibustion combined with traditional Chinese medicine application intervention. Dazhui and bilateral Feshu were selected at acupoints, and the changes of chest surface temperature of asthmatic New Zealand rabbits were monitored by infrared thermograph.
