電針不同穴位對(duì)頸部切口痛大鼠頸段背根節(jié)內(nèi)衛(wèi)星膠質(zhì)細(xì)胞活動(dòng)的影響
[Abstract]:Objective: to observe the effect of electroacupuncture on (SGCs) activity of satellite glia in (DRG) of cervical dorsal root ganglion (DRG) of rats with incision pain in thyroid region, and to explore the mechanism of relieving postoperative pain or acupuncture analgesia by acupuncture. Methods: SD rats were randomly divided into normal group, model group, FUP group, Hegu-Neiguan group and Zusanli-Yanglingquan group with 20 rats in each group. In addition to the normal group, a longitudinal incision was made along the median line of the neck of the rat and stimulated repeatedly to make the model of incision pain in the thyroid region. Each treatment group was treated with electroacupuncture at Zusanli Yangling Quan (Zusanli-Yanglingquan) once a day for 3 days. The thermal pain threshold of the incision was measured in rats. Detection of glial fibrillary acidic protein (GFAP), in rat spinal cord (C) 2-C 6DRGs by immunofluorescence, Real-time PCR,ELISA and Western blot Connexin 43 (Cx 43) immunofluorescence intensity, interleukin-1 尾 (IL-1 尾), tumor necrosis factor- 偽 (TNF- 偽), IL-6mRNA and protein expression and Cx 43 protein expression. Results: the thermal pain threshold of the model group was significantly shorter than that of the normal group (P0.05). Compared with model group and Zusanli-Yanglingquan group, the thermal pain threshold in FUP group and Hegu Neiguan group was significantly longer than that in model group and Zusanli-Yanglingquan group (P0.05) GFAP immunofluorescence intensity and mRNA expression level and IL-1 尾, TNF- 偽 were significantly increased in FUP group and Hegu Neiguan group. The expression of IL-6mRNA and protein, the immunofluorescence intensity and protein expression of Cx 43 were significantly up-regulated in the model group (P0.05). Compared with the model group, GFAP immunofluorescence intensity and mRNA expression level, IL-1 尾, TNF- 偽, IL-6mRNA expression level and Cx 43 immunofluorescence intensity and protein expression level after electroacupuncture at Hegu and Neiguan acupoints of both sides were compared, IL-1 尾, IL-1 尾 in FUT group were significantly higher than those in control group. The expression of TNF- 偽 and IL-6 protein in Hegu-Neiguan group were significantly down-regulated (P0.05), except IL-1 尾 and TNF- 偽 mRNA in Zusanli-Yanglingquan group (P0.05). Conclusion: electroacupuncture at Hegu Neiguan point can relieve the pain of neck incision in rats, which may be related to its down-regulation of SGCs activity in C2-C 6DRGs, the reduction of inflammatory cytokines release and the decrease of information exchange among SGCs.
