【摘要】：目的:建立新疆一枝蒿有效部位主要成分的快速識(shí)別和高效制備的方法,為民族藥的研究提供參考。方法:采用液相-高分辨質(zhì)譜(LC-HRMS/MS)法對(duì)一枝蒿有效部位的主要成分進(jìn)行初步推測,利用高效液相色譜、紫外光譜和質(zhì)譜法將部分化合物與相應(yīng)對(duì)照品進(jìn)行比對(duì)分析確定其名稱;對(duì)尚未確定的化合物采用柱分離及制備液相色譜技術(shù)快速獲得單體,并確定其結(jié)構(gòu)。結(jié)果:從一枝蒿有效部位分離出5個(gè)化合物,鑒定出其中2個(gè)化合物為艾黃素和紫花牡荊素;獲得1個(gè)單體化合物(得率為0.35 mg/g,純度為98.5%),經(jīng)結(jié)構(gòu)確證為6-去甲氧基-4′-O-甲基茵陳色原酮-7-O-β-D-葡萄糖苷。結(jié)論:本試驗(yàn)建立的方法實(shí)現(xiàn)了目標(biāo)成分的快速分離、鑒定、制備,可應(yīng)用于民族藥復(fù)雜的物質(zhì)基礎(chǔ)研究。
[Abstract]:Objective: to establish a method for rapid identification and efficient preparation of the main components of active parts of Artemisia annua in Xinjiang, and to provide a reference for the study of ethnic medicine. Methods: the main components of the active parts of Artemisia annua were preliminarily inferred by liquid phase high resolution mass spectrometry (LC-HRMS/MS). The names of some compounds were compared with the corresponding reference substances by HPLC, UV and MS. The undetermined compounds were separated by column and their structures were determined by liquid chromatography. Results: five compounds were isolated from the active parts of Artemisia annua and two of them were identified as Artemistein and Bauhinia. A monomer compound with a purity of 98.5% (yield of 0.35 mg/g,) was obtained. The structure of the compound was confirmed to be 6-demethoxy-4- O-methylcaptophan -7-O- 尾 -D-glucoside. Conclusion: the method established in this experiment can be used in the study of the complex material base of the national medicine, and the rapid separation, identification and preparation of the target components can be realized.
