【摘要】：目的:探討朝醫(yī)麻黃定喘湯對支氣管哮喘小鼠氣道重塑及核因子-κB(NF-κB)信號通路的影響。方法:50只清潔級雌性BABL/c小鼠,隨機分為5組,每組10只,分別為正常對照組,模型組,麻黃定喘湯低、高劑量組,地塞米松組。以卵清蛋白致敏、激發(fā)制備小鼠哮喘模型,在末次激發(fā)24h后處死。取小鼠肺泡灌洗液(BALF)及肺組織檢測炎性細胞的數(shù)量變化,白介素-4(IL-4)、白介素-5(IL-5)、白介素-13(IL-13)的水平及NF-κBp65蛋白的表達。結(jié)果:與正常對照組比較,模型組BALF中炎性細胞計數(shù)、IL-4、IL-5、IL-13水平均增高;肺組織NF-κBp65蛋白表達水平均顯著升高(P0.01)。與模型組比較,麻黃定喘湯組小鼠BALF中炎性細胞計數(shù)、IL-4、IL-5、IL-13水平及NF-κBp65蛋白表達水平均顯著降低(P0.05,P0.01),不同劑量麻黃定喘湯組間上述各指標間差異無統(tǒng)計學(xué)意義。結(jié)論:麻黃定喘湯可抑制哮喘小鼠氣道重塑的發(fā)生,其機制可能是通過抑制NF-κB信號通路而實現(xiàn)的。
[Abstract]:Aim: to investigate the effect of Chaoyi mahuang Dingchuan decoction on airway remodeling and nuclear factor-魏 B (NF- 魏 B) signaling pathway in asthmatic mice. Methods Fifty female BABL/c mice of clean grade were randomly divided into 5 groups: normal control group, model group, ephedrine Dingchuan decoction low dose group, high dose group and dexamethasone group. Mouse asthma model was induced by sensitizing ovalbumin and killed 24 hours after the last stimulation. The changes of inflammatory cells, the levels of interleukin-4 (IL-4), interleukin-5 (IL-5), interleukin-13 (IL-13) and the expression of NF- 魏 Bp65 were measured in the alveolar lavage fluid (BALF) and lung tissue of mice. Results: compared with the normal control group, the inflammatory cell count and the level of IL-5 and IL-13 in the BALF of the model group were all increased, while the expression of NF- 魏 Bp65 protein in the lung tissue was significantly increased (P0.01). Compared with the model group, the level of IL-4, IL-5, IL-13 and the expression of NF- 魏 Bp65 in the BALF of the mice in the mahuang Dingchuan decoction group were significantly decreased (P0.05, P0.01), but there was no significant difference among the above indexes in the different dosage groups. Conclusion: mahuang Dingchuan decoction can inhibit airway remodeling in asthmatic mice and its mechanism may be by inhibiting NF- 魏 B signaling pathway.
