【摘要】：目的:探索藏藥翼首草治療類風(fēng)濕性關(guān)節(jié)炎(RA)的多成分、多靶點(diǎn)、多途徑作用機(jī)制。方法:將選取的目標(biāo)化合物(即翼首草10個(gè)化學(xué)成分結(jié)構(gòu))采用相關(guān)軟件進(jìn)行數(shù)據(jù)的導(dǎo)入和存儲(chǔ),使用Pharm Mapper和Drug Bank數(shù)據(jù)庫進(jìn)行靶點(diǎn)預(yù)測和篩選,通過MAS 3.0數(shù)據(jù)庫對靶點(diǎn)進(jìn)行通路獲取并分析,最后使用Cytoscape 3.4.0軟件構(gòu)建翼首草"活性成分-靶點(diǎn)-通路"網(wǎng)絡(luò)。結(jié)果:將Pharm Mapper數(shù)據(jù)庫中獲得的靶點(diǎn)信息與Drug Bank數(shù)據(jù)庫中與炎癥相關(guān)藥物的靶點(diǎn)進(jìn)行比對,得到26個(gè)可能與翼首草治療RA相關(guān)的潛在作用靶點(diǎn),其中MAPK14、RXRA、ALB、PDE4D、VDR等靶點(diǎn)可能是翼首草活性成分治療RA的主要潛在靶點(diǎn)基因群;26個(gè)作用靶點(diǎn)通過通路注釋后得到57條作用通路,除27條與RA相關(guān)外,還涉及與內(nèi)分泌調(diào)節(jié)、免疫相關(guān)的其他30條作用通路。結(jié)論:根據(jù)網(wǎng)絡(luò)藥理學(xué)研究結(jié)果認(rèn)為,翼首草可通過作用于炎癥、免疫、內(nèi)分泌等相關(guān)的靶點(diǎn)及通路,發(fā)揮治療RA的作用。
[Abstract]:Objective: to explore the multi-component, multi-target and multi-pathway action mechanism of Rhizoma Astragalus in the treatment of rheumatoid arthritis (RA). Methods: the selected target compounds (i.e., 10 chemical composition structures of Pharm Mapper and Drug Bank) were used to import and store the data, and the target sites were predicted and screened using Pharm Mapper and Drug Bank databases. The target was acquired and analyzed by MAS 3.0 database. Finally, the "active Component-Target Pathway" network was constructed by using the software of Cytoscape 3.4.0. Results: the target information obtained from the Pharm Mapper database was compared with the targets of anti-inflammatory drugs in the Drug Bank database, and 26 potential targets related to the treatment of RA by Pharm Mapper were obtained. The target sites such as MAPK14, RXRAB, PDE4D, VDR and so on may be the main potential target gene groups for the treatment of RA by active components of Acanthopsis pterygii, and 57 action pathways are obtained by means of pathway annotation of 26 targets, including 27 related to RA, and also related to endocrine regulation. Other 30 other immune-related pathways. Conclusion: according to the results of the network pharmacological studies, it is concluded that Acanthopsis pterygii can play a role in the treatment of RA by acting on inflammatory, immunological, endocrine and other related targets and pathways.
