督脈電針對(duì)慢性腦缺血大鼠OX2R及其配體表達(dá)的影響

【摘要】:目的:研究督脈電針在慢性腦缺血時(shí)大鼠腦食欲素受體-2(OX2R)及其配體在不同時(shí)間點(diǎn)缺血后的變化。方法:實(shí)驗(yàn)中運(yùn)用分次結(jié)扎雙側(cè)頸總動(dòng)脈法建立慢性腦缺血模型,將Wistar大鼠分4組,造模成功后給予督脈電針干預(yù),免疫組織化學(xué)法觀察OX2R的表達(dá),同時(shí)運(yùn)用ELISA技術(shù)檢測(cè)各組大鼠額葉皮質(zhì)區(qū)Orexin-A和Orexin-B的總含量變化。結(jié)果:1個(gè)月、2個(gè)月時(shí),模型組OX2R及其配體表達(dá)高于正常組,同時(shí)呈遞增趨勢(shì);在相同時(shí)間進(jìn)行組間比較,1個(gè)月時(shí),模型組和針刺治療組OX2R及其配體的表達(dá)無(wú)顯著差別,立即針刺組OX2R及其配體的表達(dá)較模型組有升高;2個(gè)月時(shí)針刺治療組OX2R及其配體表達(dá)較模型組有明顯升高,而立即針刺組與針刺治療組相比OX2R及其配體表達(dá)有升高。結(jié)論:實(shí)驗(yàn)觀察到OX2R及其配體的表達(dá)在1個(gè)月和2個(gè)月時(shí)隨著時(shí)間延長(zhǎng)而升高,經(jīng)過督脈電針干預(yù)后促進(jìn)了OX2R及其配體表達(dá),同時(shí)觀察到早期干預(yù)對(duì)OX2R及其配體表達(dá)的影響優(yōu)于針刺治療組。
[Abstract]:Aim: to study the changes of orexin receptor-2 (OX2R) and its ligand after chronic cerebral ischemia in rats with du meridian electroacupuncture (EA) at different time points. Methods: the model of chronic cerebral ischemia was established by bilateral common carotid artery ligation. Wistar rats were divided into 4 groups. The rats were treated with du meridian electroacupuncture intervention and the expression of OX2R was observed by immunohistochemical method. The total contents of Orexin-A and Orexin-B in frontal cortex were detected by Elisa. Results: after 1 month and 2 months, the expression of OX2R and its ligand in model group was higher than that in normal group, and the expression of OX2R and its ligand in model group and acupuncture group had no significant difference at the same time. The expression of OX2R and its ligand in the immediate acupuncture group was higher than that in the model group, and the expression of OX2R and its ligand in the acupuncture group was significantly higher than that in the model group at 2 months, but the expression of OX2R and its ligand in the immediate acupuncture group was higher than that in the acupuncture group. Conclusion: it was observed that the expression of OX2R and its ligand increased with the prolongation of time at 1 and 2 months. The expression of OX2R and its ligand was promoted by du pulse electroacupuncture intervention. At the same time, the effect of early intervention on OX2R and its ligand expression was better than that of acupuncture group.
