PAR4、TRPV1在電針緩解腸易激綜合征大鼠內(nèi)臟痛敏中的作用研究

【摘要】:目的:觀察PAR4、TRPV1在電針緩解腸易激綜合征(irritable bowel syndrome,IBS)內(nèi)臟痛敏中的作用,探討針刺緩解大鼠內(nèi)臟痛敏的可能機(jī)制。方法:將SPF級(jí)成年雄性Wistar大鼠32只隨機(jī)分為正常對(duì)照組、模型組、電針大腸俞組、電針上巨虛組,每組8只,采用乙酸灌腸法復(fù)制IBS內(nèi)臟痛敏模型。造模成功后電針治療,每天1次,連續(xù)7 d。在電針結(jié)束后,觀察各組大鼠糞便性狀并根據(jù)糞便性狀分級(jí)進(jìn)行糞便性狀的評(píng)分,檢測(cè)糞便含水量和引起腹部回縮反射的最小容量閾值,采用western-blotting檢測(cè)大鼠結(jié)腸組織中PAR4、TRPV1蛋白的含量。結(jié)果:模型組大鼠糞便性狀評(píng)分和糞便含水量與正常對(duì)照組相比明顯增加(P0.01,P0.01),電針大腸俞組、電針上巨虛組大鼠糞便性狀評(píng)分(P0.01,P0.01)和糞便含水量降低(P0.05,P0.05)。模型組與正常對(duì)照組相比引起大鼠腹部回縮反射的容量閾值明顯降低(P0.01),電針大腸俞和電針上巨虛后引起大鼠腹部回縮反射的容量閾值顯著增加(P0.01,P0.01);模型組結(jié)腸組織PAR4蛋白的表達(dá)水平與正常對(duì)照組相比有升高的趨勢(shì),但差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05);與模型組相比電針上巨虛組結(jié)腸組織PAR4蛋白表達(dá)水平下降,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。模型組結(jié)腸組織TRPV1蛋白的表達(dá)水平與正常對(duì)照組相比升高(P0.05)。與模型組相比電針大腸俞組、電針上巨虛組結(jié)腸組織TRPV1蛋白表達(dá)水平有下降的趨勢(shì)。結(jié)論:電針可以有效緩解IBS內(nèi)臟痛敏,其機(jī)制可能和調(diào)控IBS內(nèi)臟痛敏大鼠結(jié)腸PAR4、TRPV1蛋白的表達(dá)有關(guān)。
[Abstract]:Objective: to observe the effect of PAR4 / TRPV1 on relieving visceral pain sensitivity of irritable bowel syndrome (IBSs) of irritable bowel syndrome (IBS), and to explore the possible mechanism of relieving visceral pain sensitivity by acupuncture. Methods: Thirty-two SPF grade adult male Wistar rats were randomly divided into three groups: normal control group, model group, electroacupuncture group (Dachangshu group) and Shangzhouxu group (8 rats in each group). IBS visceral pain sensitive model was induced by acetic acid enema. The model was successfully treated with electroacupuncture once a day for 7 days. After electroacupuncture, the fecal traits of rats in each group were observed and graded according to fecal traits, the fecal water content and the minimum volume threshold of abdominal reflex were measured. Western-blotting was used to detect the protein content of PAR4 and TRPV1 in rat colon. Results: the scores of fecal traits and fecal water content in the model group were significantly higher than those in the normal control group, and the scores of fecal traits were significantly increased in the Dachang Shu group and the Shangzhouxu group (P 0.01 P 0.01) and the fecal water content was decreased (P 0.05 P 0.05). Compared with the normal control group, the volume threshold of abdominal reflex in the model group was significantly lower than that in the normal control group, and that in the model group was significantly higher than that in the model group. Compared with the normal control group, the expression of PAR4 protein increased. Compared with the model group, the expression of PAR4 protein in the colon tissue of the model group decreased, and the difference was statistically significant (P 0.05). The expression of TRPV1 protein in colonic tissue of model group was higher than that of normal control group (P 0.05). Compared with the model group, the expression of TRPV1 protein in the colon tissue of the Shangjiu asthenia group showed a decreasing trend compared with the model group. Conclusion: electroacupuncture can effectively relieve visceral pain sensitivity of IBS, and its mechanism may be related to regulating the expression of TRPV1 protein in colon of IBS visceral pain sensitive rats.
