哈尼族驗(yàn)方“莫那奇”對(duì)腦出血大鼠腦組織BDNF表達(dá)影響的研究

【摘要】:研究目的通過(guò)對(duì)腦源性神經(jīng)營(yíng)養(yǎng)因子(BDNF)表達(dá)水平等指標(biāo)的檢測(cè),研究哈尼族驗(yàn)方“莫那奇”對(duì)腦出血的治療作用,探討“莫那奇”在腦出血治療中的作用機(jī)制。實(shí)驗(yàn)方法將72只SD大鼠隨機(jī)分為空白組、假手術(shù)組、模型組和莫那奇組,采用尾狀核自體血注入法復(fù)制大鼠腦出血模型,在造模3小時(shí)后及“莫那奇”干預(yù)后第3天按照Berderson行為學(xué)評(píng)分法對(duì)大鼠神經(jīng)缺損體征進(jìn)行評(píng)分并處死取材,應(yīng)用干濕重法測(cè)定其腦組織含水量、HE染色法觀察腦組織形態(tài)學(xué)改變情況及用原位雜交方法檢測(cè)干預(yù)后BDNF陽(yáng)性細(xì)胞數(shù)量的變化。實(shí)驗(yàn)結(jié)果1.腦出血3小時(shí)后,模型組和莫那奇組均表現(xiàn)為出血對(duì)側(cè)肢體癱瘓,空白組和假手術(shù)組大鼠基本無(wú)神經(jīng)功能異常表現(xiàn)。經(jīng)“莫那奇”干預(yù)3天后大鼠神經(jīng)缺損體征有所改善,與模型組及治療前相比,差異具有統(tǒng)計(jì)學(xué)意義(P0.05)。2.干濕重法結(jié)果顯示,模型組和莫那奇組腦組織含水量較空白組和假手術(shù)組顯著升高(P0.01),其中應(yīng)用“莫那奇”治療后大鼠組腦組織含水量較模型組降低(P0.05)。3.HE染色顯示,模型組大鼠腦組織出血灶及周?chē)?xì)胞大多壞死變性,胞質(zhì)內(nèi)有較多含鐵血黃素顆粒,細(xì)胞核濃縮,染色質(zhì)崩解、分散于胞漿內(nèi),炎性細(xì)胞浸潤(rùn),膠質(zhì)細(xì)胞形成纖維囊腫,散在于血腫周?chē)?莫那奇組與模型組相比細(xì)胞結(jié)構(gòu)較規(guī)整,排列較整齊,炎性細(xì)胞較少,空白組和假手術(shù)組組織及細(xì)胞結(jié)構(gòu)完整,無(wú)明顯病理改變。4.原位雜交結(jié)果顯示莫那奇組BDNF陽(yáng)性細(xì)胞數(shù)顯著多于模型組(P0.01),空白組和假手術(shù)組基本無(wú)BDNF陽(yáng)性細(xì)胞。結(jié)論實(shí)驗(yàn)證實(shí)哈尼族驗(yàn)方“莫那奇”能夠改善大鼠神經(jīng)缺損體征、降低腦組織含水量、保護(hù)腦組織及神經(jīng)元的形態(tài)結(jié)構(gòu)、促進(jìn)腦出血大鼠腦內(nèi)腦源性神經(jīng)營(yíng)養(yǎng)因子BDNF的表達(dá),對(duì)腦出血后的腦組織損傷發(fā)揮重要的保護(hù)作用。
[Abstract]:Objective to study the therapeutic effect of Monaci on intracerebral hemorrhage (ICH) by detecting the expression level of BDNF, and to explore the mechanism of Monachi in the treatment of intracerebral hemorrhage (ICH). Methods Seventy-two Sprague-Dawley rats were randomly divided into three groups: blank group, sham operation group, model group and monazide group. The rat model of intracerebral hemorrhage was induced by autologous blood injection of caudate nucleus. After 3 hours of model making and 3 days after "Monachy" intervention, the rats were scored according to the Berderson behavioral score method, and the rats were killed to take the materials. The changes of brain tissue morphology and the number of BDNF positive cells after intervention were observed by using dry and wet weight method and HE staining. Experimental results 1. After 3 hours of intracerebral hemorrhage, the model group and Monachy group showed the contralateral limb paralysis, while the blank group and the sham operation group showed no abnormal neurological function. The physical signs of nerve defect in rats were improved after 3 days of "monagi" intervention, and the difference was statistically significant compared with the model group and before treatment (P0.05. 2). The results of dry and wet weight method showed that the water content of brain tissue in the model group and monazi group was significantly higher than that in the blank group and sham-operation group, and the water content of brain tissue in the model group was lower than that in the model group after the treatment of "monagi". 3. The results of HE staining showed that the water content of brain tissue in the model group was lower than that in the model group. In the model group, most of the cerebral hemorrhage foci and peripheral cells were necrotic and denatured, and there were more hemosiderin granules, nuclear concentration, chromatin disintegration, dispersion in the cytoplasm, infiltration of inflammatory cells and formation of fibrocysts in glial cells in the model group. Compared with the model group, the monaqi group had more regular cell structure, neatly arranged and fewer inflammatory cells. The tissue and cell structure of the blank group and the sham operation group were intact, and there were no obvious pathological changes. 4. The results of in situ hybridization showed that the number of BDNF positive cells in Monachi group was significantly higher than that in model group (P 0.01), and there were no BDNF positive cells in blank group and sham operation group. Conclusion Monaci can improve the signs of nerve defect, decrease the water content of brain tissue, protect the morphologic structure of brain tissue and neuron, and promote the expression of brain-derived neurotrophic factor (BDNF) in the brain of rats with intracerebral hemorrhage. It plays an important role in protecting brain tissue injury after intracerebral hemorrhage.
