電針井穴對血管性癡呆大鼠海馬CA1區(qū)p38MAPK的表達及細胞凋亡的影響

【摘要】:目的:觀察電針井穴對血管性血管性癡呆(VD)模型大鼠學(xué)習(xí)記憶能力及海馬CA1區(qū)p38絲裂原活化蛋白激酶(p38MAPK)表達及細胞凋亡的影響,探討電針井穴治療VD的可能作用機制,為臨床上針刺井穴治療VD提供部分實驗及理論依據(jù)。方法:1.選擇清潔級成年雄性Wistar大鼠70只,體重(275±25)g,經(jīng)1周適應(yīng)性飼養(yǎng)后,為減少動物回避性反應(yīng)差異造成的差異,用跳臺實驗篩除特別敏感和過于遲鈍的大鼠,保留60只,電腦隨機數(shù)字法選取假手術(shù)組8只,剩余大鼠根據(jù)改良的四血管阻斷法(4-VO)制備VD模型,模型復(fù)制后4天,依據(jù)跳臺實驗及神經(jīng)行為學(xué)評分判定模型是否復(fù)制成功,電腦隨機數(shù)字法分為電針組、西藥組、模型對照組3組,各8只。術(shù)后10天,電針組予以電針治療,每天1次,10天為1個療程,共3個療程,西藥組予以吡拉西坦灌胃治療,每天1次,療程與針刺組平行,模型對照組與假手術(shù)組常規(guī)飼養(yǎng),不做特殊處理。2.療程結(jié)束后先對各組大鼠進行跳臺實驗及神經(jīng)行為學(xué)評分測定,然后取材,采用免疫組化法檢測各組大鼠海馬CA1區(qū)p38MAPK表達,Tunel法檢測各組大鼠海馬CA1區(qū)細胞凋亡。采用SPSS17.0對實驗數(shù)據(jù)進行統(tǒng)計分析。結(jié)果:1.模型復(fù)制后第4d,跳臺實驗檢測潛伏期、錯誤次數(shù),同時檢測神經(jīng)行為學(xué)評分。結(jié)果:模型復(fù)制組與假手術(shù)組相比潛伏期降低(P0.01),錯誤次數(shù)增多(P0.05),神經(jīng)行為學(xué)評分增高(P0.01),表明模型復(fù)制成功。2.療程結(jié)束后,再次檢測各組大鼠跳臺實驗潛伏期、錯誤次數(shù)及神經(jīng)行為學(xué)評分。結(jié)果:結(jié)果模型對照組與電針組相比潛伏期降低(P0.01),錯誤次數(shù)增多(P0.05),神經(jīng)行為學(xué)評分增高(P0.05),差距有統(tǒng)計學(xué)意義;模型對照組與西藥組相比潛伏期降低(P0.05),差異有統(tǒng)計學(xué)意義,錯誤次數(shù)增多(P0.05),神經(jīng)行為學(xué)評分(P0.05),差異無統(tǒng)計學(xué)意義,電針組與西藥組相比潛伏期延長(P0.05),錯誤次數(shù)減少(P0.05),神經(jīng)行為學(xué)評分(P0.05),差異無統(tǒng)計學(xué)意義。結(jié)果表明電針組能明顯改善VD大鼠的學(xué)習(xí)記憶能力、神經(jīng)行為學(xué)評分,且電針組在改善學(xué)習(xí)記憶能力方面優(yōu)與西藥組。3.治療結(jié)束后,免疫組化法檢測各組大鼠海馬CA1區(qū)p38MAPK表達。與模型對照組相比,假手術(shù)組平均灰度值增高(P0.01),陽性細胞表達減少;模型對照組與西藥組相比,平均灰度值降低(P0.01),差異有統(tǒng)計學(xué)意義,與電針組相比平均灰度值減少(P0.01),陽性細胞表達增多;西藥組與電針組相比,灰度值(P0.05),差異無統(tǒng)計學(xué)意義;電針組與假手術(shù)組相比,平均灰度值增高(P0.05),結(jié)果表明電針組能降低海馬CA1區(qū)p38MAPK的陽性表達。4.Tunel法檢測海馬CA1區(qū)細胞凋亡,假手術(shù)組與模型對照組相比,細胞凋亡指數(shù)明顯降低(P0.01),差異有統(tǒng)計學(xué)意義,凋亡程度明顯減輕;電針組與模型對照組相比,細胞凋亡程度較輕(P0.01),差異有統(tǒng)計學(xué)意義;西藥組與模型對照組比較,細胞凋亡程度減輕(P0.01),差異有統(tǒng)計學(xué)意義;電針組與假手術(shù)相比,細胞凋亡指數(shù)增高(P0.01),差異有統(tǒng)計學(xué)意義,西藥組與假手術(shù)組相比,細胞凋亡指數(shù)增高(P0.01),差異有統(tǒng)計學(xué)意義。西藥組與電針組相比,細胞凋亡指數(shù)增高(P0.01),差異有統(tǒng)計學(xué)意義,結(jié)果表明電針井穴能降低海馬CA1區(qū)細胞凋亡指數(shù),減少神經(jīng)元細胞凋亡,且療效優(yōu)于西藥組。結(jié)論:1.電針井穴能改善VD大鼠學(xué)習(xí)記憶能力,降低VD大鼠神經(jīng)行為學(xué)評分。2.電針井穴能抑制p38MAPK的表達,提示電針井穴治療VD可能是通過抑制p38MAPK信號通道,減輕神經(jīng)元細胞凋亡。3.電針井穴能減輕VD大鼠海馬CA1區(qū)細胞凋亡,減輕繼發(fā)性損傷,促進神經(jīng)元的修復(fù)。
[Abstract]:Objective: To observe the effects of Electroacupuncture on the learning and memory ability of vascular dementia (VD) model rats and the expression of p38 mitogen activated protein kinase (p38MAPK) and apoptosis in hippocampus CA1 region, and to explore the possible mechanism of the treatment of VD by Electroacupuncture well points, and to provide some experimental and theoretical basis for the treatment of VD in clinical acupuncture points. Method: 1. 70 adult male Wistar rats were selected, weight (275 + 25) g, after 1 weeks of adaptive feeding, to reduce the difference of the difference in the difference of animal avoidance response. 60 rats were retained, 8 rats were selected by the random number method of computer, and the remaining rats were selected according to the improved four vascular blocking method (4- VO) VD model was prepared. 4 days after the replicating of the model, the model was made to determine whether the model was replicated successfully. The computer random number method was divided into the electroacupuncture group, the western medicine group and the model control group, each 8. The positive expression of p38MAPK in the hippocampal CA1 area was reduced by the positive electroacupuncture group. The apoptosis index of the hippocampal CA1 region was detected by.4.Tunel method. Compared with the model control group, the apoptosis index decreased significantly (P0.01), the difference was statistically significant and the degree of apoptosis was significantly reduced. Compared with the model control group, the apoptosis degree was less than that of the model control group (P0.01), and there were differences in the cell apoptosis degree (P0.01). Statistical significance: compared with the model control group, the degree of apoptosis was reduced (P0.01) in the western medicine group and the difference was statistically significant. The apoptosis index increased (P0.01) in the electroacupuncture group compared with the sham operation (P0.01), and the difference was statistically significant in the western medicine group and the sham operation group (P0.01), the difference was statistically significant. Compared to the group, the apoptosis index increased (P0.01), the difference was statistically significant. The results showed that the electroacupuncture hole can reduce the apoptosis index of CA1 area in the hippocampus and reduce the apoptosis of neuron cells, and the effect is better than that of the western medicine group. The expression of p38MAPK suggests that the treatment of VD by Electroacupuncture well points may be by inhibiting the p38MAPK signal channel and reducing the apoptosis of.3. electroacupuncture at the neuronal cell apoptosis, reducing the apoptosis in the CA1 region of the hippocampus of VD rats, reducing secondary damage and promoting the repair of neurons.
