【摘要】：目的探討電針干預(yù)對布比卡因排尿功能障礙的影響及相關(guān)作用機(jī)制。方法選取6周齡60只清潔級SD雄性大鼠分為對照組、模型組和預(yù)處理組,每組各20只。模型組和預(yù)處理組均經(jīng)腰麻注入建立布比卡因中毒大鼠模型。建模前模型組大鼠接受電針預(yù)處理。測定和記錄各組大鼠膀胱測壓參數(shù)。免疫染色組化法測定和比較各組大鼠逼尿肌細(xì)胞凋亡率和細(xì)胞bax、bcl-2表達(dá),Western blot法測定和比較各組大鼠脊髓BDNF、Trk B和P2X3蛋白表達(dá)。結(jié)果對照組和預(yù)處理組大鼠單日排尿量、殘余尿量和膀胱容量均顯著高于模型組大鼠(P0.05),膀胱內(nèi)壓均顯著低于模型組大鼠(P0.05)。對照組和預(yù)處理組大鼠逼尿肌細(xì)胞AI值均顯著低于模型組(P0.01)。對照組和預(yù)處理組大鼠逼尿肌細(xì)胞bax表達(dá)顯著高于模型組(P0.01),逼尿肌細(xì)胞bcl-2表達(dá)顯著低于模型組(P0.01)。對照組和預(yù)處理組大鼠脊髓BDNF、Trk B表達(dá)顯著低于模型組(P0.01),脊髓P2X3蛋白表達(dá)顯著高于模型組(P0.01)。結(jié)論電針預(yù)處理可以改善布比卡因中毒大鼠排尿功能障礙,其作用機(jī)制可能與調(diào)控膀胱逼尿肌Bcl-2/Bax及脊髓BDNF、Trk B、P2X3蛋白表達(dá)相關(guān)。
[Abstract]:Objective to investigate the effect of electroacupuncture intervention on urinary dysfunction of bupivacaine and its related mechanism. Methods Sixty clean SD male rats aged 6 weeks were divided into control group, model group and pretreatment group with 20 rats in each group. The model group and pretreatment group were injected with spinal anesthesia to establish the model of bupivacaine poisoning in rats. Rats in the model group were pretreated with electroacupuncture before modeling. The parameters of bladder manometry were measured and recorded in each group. The apoptosis rate of detrusor muscle cells and the expression of bcl-2 in the cells of detrusor muscle were detected and compared by immunohistochemical method. The expression of BDNF Trk B and P2X3 protein in spinal cord of rats in each group was detected and compared by Western blot method. Results the urine volume, residual urine volume and bladder volume in the control group and pretreatment group were significantly higher than those in the model group, and the intravesical pressure was significantly lower than that in the model group. The AI of detrusor cells in control group and preconditioning group was significantly lower than that in model group (P 0.01). The bax expression of detrusor cells in the control group and preconditioning group was significantly higher than that in the model group, and the bcl-2 expression in the detrusor cells was significantly lower than that in the model group. The expression of BDNF Trk B in the spinal cord of the control group and preconditioning group was significantly lower than that of the model group, and the expression of P2X3 protein in the spinal cord was significantly higher than that in the model group. Conclusion electroacupuncture preconditioning can improve urination dysfunction in rats with bupivacaine poisoning, and its mechanism may be related to the regulation of Bcl-2/Bax in bladder detrusor muscle and BDNF Trk BP2X3 protein expression in spinal cord.
