基于內(nèi)皮損傷學(xué)說的冠心病中藥干預(yù)研究

【摘要】:目的:本研究依據(jù)“內(nèi)皮損傷學(xué)說”,通過血清藥理學(xué)實(shí)驗(yàn),探究中藥對內(nèi)皮細(xì)胞和級聯(lián)反應(yīng)細(xì)胞干擾素的影響,為科研與臨床驗(yàn)證中藥治療冠心病療效提供候選檢測方法。方法:1選取健康受試者10例,臨床劑量靜點(diǎn)舒血寧注射液,進(jìn)行安全性臨床觀察,同時(shí)制備含藥血清。2制作內(nèi)皮細(xì)胞PAF損傷模型,以空白和含藥血清培養(yǎng)細(xì)胞,觀察細(xì)胞形態(tài)與增殖,以Elisa試劑盒檢測細(xì)胞上清分泌物中NO、VE-Ca、ICAM-1、VCAM-1濃度,觀察舒血寧對細(xì)胞及級聯(lián)反應(yīng)細(xì)胞干擾因素的影響,研究其對冠心病內(nèi)皮損傷的干預(yù)作用。結(jié)果:1舒血寧注射液臨床試驗(yàn)觀察,受試者前后生命體征、肝腎功、血常規(guī)、凝血五項(xiàng)中,體溫、淋巴細(xì)胞、中性粒細(xì)胞P值0.05,考慮為樣本量過少所致;余項(xiàng)P值均0.05,表明用藥前后無明顯變化。不良反應(yīng)1例,1級輕度,可能與試驗(yàn)藥物無關(guān)。2造模:當(dāng)以濃度為25umol/L的PAF作用4h后,細(xì)胞存活率平均值為47.45±6.95,細(xì)胞損傷率近50%,考慮為理想造模條件。細(xì)胞增殖隨PAF濃度的升高、誘導(dǎo)時(shí)間的延長而受到抑制程度也加重,PAF誘導(dǎo)細(xì)胞損傷。3與模型對照組相比,含藥血清組細(xì)胞形態(tài)生長狀態(tài)良好,增殖72.9%±4.87%,NO濃度下降,VE-Ca、ICAM-1、VCAM-1濃度升高,與空白對照組相近,與模型對照組存在明顯差異。結(jié)論1舒血寧注射液能夠通過改善細(xì)胞形態(tài),促進(jìn)細(xì)胞增殖,調(diào)節(jié)細(xì)胞分泌功能,對內(nèi)皮細(xì)胞和細(xì)胞級聯(lián)反應(yīng)干擾素產(chǎn)生影響。2以人體血清及細(xì)胞實(shí)驗(yàn)為依托的中藥治療冠心病療效的候選檢測方法具有可行性。
[Abstract]:Objective: To investigate the effect of Chinese medicine on the interferon in endothelial cells and cascade reaction cells through the theory of "endothelial injury", and to provide a candidate detection method for the therapeutic effect of Chinese medicine in the treatment of coronary heart disease by scientific research and clinical validation. 1 the clinical dose of Shuxuening injection was carried out in 10 healthy subjects. The PAF damage model of endothelial cells was prepared by the preparation of.2 containing serum, and the cell morphology and proliferation were observed with blank and drug containing serum. The concentration of NO, VE-Ca, ICAM-1 and VCAM-1 in the secretions of cell supernatant was detected by Elisa kit, and the effects of Shuxuening on the cell and cascade reaction cells were observed. The interventional effect of endothelium injury in coronary heart disease. Results: 1 the clinical trial of Shuxuening injection was observed in the clinical trial. In the subjects, the body temperature, the liver and kidney work, the blood routine and the coagulation five, the body temperature, the lymphocyte and the P value of neutrophils were 0.05, and the value of the remainder P was 0.05, indicating that there were no obvious changes before and after the drug use, 1 cases of adverse reactions and 1 light weight. Conclusion 1 Shuxuening Injection can improve the cell morphology, promote the cell proliferation, regulate the cell secretion function, and the endothelial cells and the endothelial cells. Cell cascade reaction interferon production affects.2. It is feasible to detect human coronary heart disease with Chinese medicine based on human serum and cell experiments.
