苗族藥了哥王不同提取物對正常大鼠肝毒性的影響

【摘要】:目的:考察了哥王不同提取物對正常大鼠肝毒性的影響,并探討其作用機制。方法:選用SD大鼠72只,隨機分成空白組、乙醇提取物組、石油醚提取物組、乙酸乙酯提取物組、正丁醇提取物組和水部位提取物組,每組12只。不同給藥組連續(xù)灌胃給藥2周,每隔2日稱量大鼠體重,于第15天,每組隨機取6只,麻醉后腹主動脈取血,離心血液,取血清測定丙氨酸氨基轉(zhuǎn)移酶(ALT),天門冬氨酸氨基轉(zhuǎn)移酶(AST),堿性磷酸酶(ALP),總膽紅素(T-BIL),總蛋白(TP),白蛋白(ALB),血糖(GLU),總膽固醇(CHOL),肌酐(Cr)。取肝組織,稱其質(zhì)量,計算臟器系數(shù),并留取肝組織作病理學檢查。每組余下的6只大鼠于第15天停止給藥,正常供給飼料與水,恢復2周,觀察大鼠恢復情況,于第28天進行上述相同指標的檢測。結(jié)果:給藥2周后,與空白組比較,乙醇提取物組和乙酸乙酯提取物組大鼠的肝臟系數(shù)明顯增大(P0.05)。乙醇提取物組血清中ALT,AST,GLB明顯升高,白蛋白/球蛋白(A/G),TP和ALB明顯降低(P0.05);乙酸乙酯提取物組血清中ALP,T-BIL,GLU明顯升高,A/G,TP和ALB明顯降低(P0.05);石油醚提取物組血清中AST,AST/ALT,ALP,GLU明顯升高,T-BIL,A/G,TP和ALB明顯降低(P0.05,P0.01);正丁醇提取物組血清中AST/ALT,T-BIL明顯升高,A/G明顯降低(P0.05,P0.01);水部位提取物組血清中A/G明顯降低(P0.05);乙醇提取物組、乙酸乙酯提取物組和石油醚提取物組大鼠肝組織能觀察到不同程度的炎細胞浸潤,出現(xiàn)一定的肝細胞脂肪變性和局灶性肝細胞壞死,其他組少見。停藥恢復2周后,各組肝臟系數(shù)均接近于空白組;各組血清中肝功能生化指標基本恢復到空白組水平;各組大鼠肝組織細胞結(jié)構(gòu)清晰,少見炎細胞浸潤,未見脂肪變性和肝細胞壞死。結(jié)論:苗族藥了哥王乙醇提取物組、乙酸乙酯提取物組對肝損傷較大,具有明顯的肝毒性;石油醚提取物組對肝也有一定的損傷,但比乙醇提取物組和乙酸乙酯提取物組損傷程度小;正丁醇提取物組毒性較小,水部位提取物組無毒性;停藥2周恢復后,各組大鼠反映肝功能的生化指標基本恢復到空白組水平,大鼠的肝損傷明顯的減輕,表明了哥王的肝毒性具有一定的可逆性。
[Abstract]:Aim: to investigate the effects of different extracts of Koking on the hepatotoxicity of normal rats and its mechanism.Methods: Seventy-two SD rats were randomly divided into control group, ethanol extract group, petroleum ether extract group, ethyl acetate extract group, n-butanol extract group and water extract group with 12 rats in each group.Rats in different groups were given intragastric administration for 2 weeks and weighed every 2 days. On the 15th day, 6 rats in each group were randomly selected. Blood was taken from abdominal aorta after anesthesia and blood was centrifuged.Serum levels of alanine aminotransferase (alt), aspartate aminotransferase (AST), alkaline phosphatase (ALP), total bilirubin (T-BILN), total protein (TP), albumin (Alb), blood glucose (GLU), total cholesterol (TC) and creatinine (Cr) were determined.Take the liver tissue, call its mass, calculate the organ coefficient, and leave the liver tissue for pathological examination.The remaining 6 rats in each group were stopped on the 15th day, fed with normal feed and water, and recovered for 2 weeks. The recovery of the rats was observed, and the same indexes were detected on the 28th day.Results: after 2 weeks of administration, compared with the blank group, the liver coefficient of the ethanol extract group and the ethyl acetate extract group increased significantly (P 0.05).In the ethanol extract group, the serum ALT AST GLB was significantly higher than that in the control group.Albumin / globulin A / G (TP) and ALB decreased significantly (P 0.05N), serum ALP T-BILN GLU in ethyl acetate extract group increased significantly (P 0.05) and ALB decreased significantly (P 0.05); serum AST / T (alt) ALPGLU significantly increased in petroleum ether extract group (P < 0.05); blood P0.05T (P 0.05) was decreased in butanol extract group (P < 0.05); blood P0.05P was significantly decreased in ethyl acetate extract group (P < 0.05); serum level of ALB was significantly decreased in butanol extract group (P < 0.01); serum level of ASTT / T (alt) was significantly increased in petroleum ether extract group (P < 0.05).In Qingzhong, AST / alt T-BIL was significantly increased and A-1 / G decreased significantly (P / P 0.05), P / G was significantly decreased in water extract group, and P / G was significantly decreased in water extract group; ethanol extract group was treated with ethanol extract.In the ethyl acetate extract group and the petroleum ether extract group, different degrees of inflammatory cell infiltration were observed in the liver tissue, and some fatty degeneration of liver cells and focal hepatocyte necrosis were observed in the other groups, which were rare in other groups.After 2 weeks of withdrawal, the liver coefficient of each group was close to that of the blank group; the biochemical indexes of liver function in the serum of each group returned to the level of the blank group; the structure of liver tissue cells in each group was clear, and the infiltration of inflammatory cells was rare.There was no fatty degeneration and hepatocyte necrosis.Conclusion: in the Miao nationality, the alcohol extract group of Koking was found to have great liver injury and obvious hepatotoxicity in ethyl acetate extract group, and the petroleum ether extract group also had some damage to the liver.The toxicity of n-butanol extract group was less than that of ethanol extract group and ethyl acetate extract group, the water extract group had no toxicity, and after 2 weeks of withdrawal, the toxicity of n-butanol extract group was less than that of ethyl acetate extract group.The biochemical indexes reflecting liver function in each group were basically restored to the level of blank group, and the liver injury of rats was obviously alleviated, indicating that the hepatotoxicity of elder brother Wang had certain reversibility.
