針刺結(jié)合益氣祛風(fēng)湯對(duì)慢性ACD小鼠病變組織中Fas及FasL表達(dá)的影響

【摘要】:目的:建立慢性ACD小鼠模型,并用針刺結(jié)合益氣祛風(fēng)湯的方法對(duì)慢性ACD模型小鼠進(jìn)行治療,進(jìn)而觀察其對(duì)小鼠組織形態(tài)學(xué)的影響,并探討其對(duì)慢性ACD模型小鼠耳病變皮膚組織中Fas及Fas L表達(dá)的影響,為臨床能夠更好地應(yīng)用針刺結(jié)合的方法治療變應(yīng)性接觸性皮炎提供可靠而客觀的依據(jù)。材料和方法:1.實(shí)驗(yàn)材料(1)實(shí)驗(yàn)動(dòng)物:60只健康的雄性昆明種小鼠。(2)實(shí)驗(yàn)用藥:通過灌胃給藥方式,中藥組給予益氣祛風(fēng)湯顆粒劑懸濁液;西藥組給予氯雷他定片懸濁液;針?biāo)幗M給予益氣祛風(fēng)湯顆粒劑懸濁液;空白組及模型組均給予等量蒸餾水。2.研究方法(1)造模方法:采用局部皮膚涂抹2,4-二硝基氯苯(2,4-DNCB)的方法制備小鼠的慢性ACD動(dòng)物模型。(2)通過觀察針刺結(jié)合益氣祛風(fēng)湯對(duì)慢性ACD模型小鼠病變耳重量差的影響,進(jìn)而評(píng)價(jià)其對(duì)慢性ACD小鼠的抑制作用。(3)采用免疫組化的方法,進(jìn)行對(duì)慢性ACD模型小鼠病變皮膚組織中Fas/Fas L的含量進(jìn)行檢測(cè),研究其表達(dá)的變化,進(jìn)一步探究針刺結(jié)合益氣祛風(fēng)湯對(duì)慢性ACD小鼠的抑制作用。結(jié)果:1.針刺結(jié)合益氣祛風(fēng)湯可明顯下調(diào)慢性ACD模型小鼠病變耳的重量差值:慢性ACD模型組的小鼠耳重量差值明顯高于空白組(P0.01),針?biāo)幗M及氯雷他定組的耳重量差值明顯低于模型組(P0.05),針刺及中藥組的耳重量差值與模型組相比均無顯著性差異(P0.05);針?biāo)幗M與氯雷他定組之間無顯著性差異(P0.05)。2.針刺結(jié)合益氣祛風(fēng)湯可明顯上調(diào)慢性ACD模型小鼠耳病變皮膚組織中Fas/Fas L的表達(dá):慢性ACD模型組的小鼠耳病變皮膚組織中的Fas、Fas L的表達(dá)均增高;針?biāo)幗M、針刺組及中藥組小鼠耳病變皮膚組織中Fas、Fas L的表達(dá)均明顯上調(diào);氯雷他定組對(duì)小鼠耳病變皮膚組織中Fas、Fas L的表達(dá)無明顯影響,且針刺結(jié)合益氣祛風(fēng)湯組與氯雷他定組之間無明顯差異。結(jié)論:1.針刺結(jié)合益氣祛風(fēng)湯對(duì)慢性ACD模型小鼠有抑制作用,且能降低其病變耳的重量差值;2.針刺結(jié)合益氣祛風(fēng)湯能明顯上調(diào)慢性ACD模型小鼠的耳病變皮膚組織中Fas和Fas L的表達(dá)。
[Abstract]:Objective: to establish the chronic ACD mouse model and treat the chronic ACD model mice with acupuncture combined with Yiqi Qufeng decoction, and then observe its effect on the histomorphology of the mice. To investigate the effect of Fas and Fas L on the expression of Fas and Fas L in the skin of chronic ACD model mice. To provide a reliable and objective basis for clinical application of acupuncture combined with acupuncture in the treatment of allergic contact dermatitis. Materials and methods: 1) Experimental animals: 60 healthy male Kunming mice. By gavage, The traditional Chinese medicine group was treated with Yiqi Qufeng decoction granule suspension; the western medicine group with loratadine tablet suspension liquid; the acupuncture and medicine group with Yiqi Qufeng decoction granule suspension; The blank group and the model group were given the same amount of distilled water. 2. The research method was 1) the method of making model: the chronic ACD animal model of mice was made by using local skin smearing with 2NCB-dinitrochlorobenzene 24-DNCB.) by observing acupuncture combined with Yiqi Qufeng decoction, the animal model of chronic ACD in mice was established by observing acupuncture combined with Yiqi Qufeng decoction. The effect on the weight difference of diseased ears in chronic ACD model mice, To evaluate its inhibitory effect on chronic ACD mice, immunohistochemical method was used to detect the content of Fas/Fas L in the diseased skin tissue of chronic ACD model mice, and to study the changes of its expression. To further explore the inhibitory effect of acupuncture combined with Yiqi Qufeng decoction on chronic ACD mice. Results 1. Acupuncture combined with Yiqi Qufeng decoction could significantly decrease the weight difference of diseased ears in chronic ACD model mice: ear weight of chronic ACD model group. The volume difference was significantly higher than that of the blank group (P 0.01), the difference of ear weight between the acupuncture group and the loratadine group was significantly lower than that of the model group (P 0.05), and there was no significant difference in the ear weight difference between the acupuncture group and the traditional Chinese medicine group compared with the model group (P 0.05), and the difference between the acupuncture group and the loratadine group was not significant (P 0.05). There was no significant difference between them. Acupuncture combined with Yiqi Qufeng decoction could obviously upregulate the expression of Fas/Fas L in the ear lesion skin of chronic ACD model mice. The expression of Fas-Fas L in the ear lesion skin tissue of chronic ACD model group was all increased. The expression of Fas-Fas L in ear lesion skin of mice in acupuncture group, acupuncture group and traditional Chinese medicine group were all upregulated obviously, while the expression of Fas-Fas L in mouse ear lesion skin tissue was not affected by loratadine group. There was no significant difference between acupuncture combined with Yiqi Qufeng decoction group and loratadine group. Conclusion: 1. Acupuncture combined with Yiqi Qufeng decoction can inhibit chronic ACD model mice. Acupuncture combined with Yiqi Qufeng decoction could obviously upregulate the expression of Fas and Fas L in the skin of chronic ACD model mice.
