地格達(dá)-4對幽門結(jié)扎性肝損傷大鼠PXR、CAR表達(dá)的影響

【摘要】:目的:研究地格達(dá)-4能否通過孕烷X受體(PXR)、組成型雄甾烷受體(CAR)表達(dá)變化治療幽門結(jié)扎性肝損傷。方法:取50只大鼠,分為5組,即正常組、模型組、地格達(dá)-4的3個劑量(1.86、3.71、7.43g/kg)組,通過幽門結(jié)扎法建立大鼠肝損傷動物模型,灌胃給藥共15d,采用實(shí)時熒光定量鏈?zhǔn)骄酆厦阜磻?yīng)(RTPCR)、蛋白印跡(Western blot)法檢測大鼠肝臟組織中PXR、CAR表達(dá)變化及測定血清和肝組織中肝損傷敏感性指標(biāo)。結(jié)果:與正常組比較,模型組血清AST、ALT、BILD2、BILT3、TBA顯著增加(P0.01,P0.05),肝組織GSH-PX顯著降低(P0.01);腹腔觀察,腹膜具有明顯的黃色、腹腔積水、黃尿、肉眼觀察肝臟淤血;形態(tài)學(xué)觀察,肝細(xì)胞有明顯的脂肪變、水腫、邊膜外肝細(xì)胞大量壞死、肝細(xì)胞具有一定程度的凋亡;與模型組比較,各給藥組對AST、ALT、BILD2、BILT3、TBA顯著降低(P0.01,P0.05),GSH-PX顯著增高(P0.01);腹腔及肝組織形態(tài)具有一定好轉(zhuǎn);RT-PCR結(jié)果,地格達(dá)-4低、中、高組對核受體CAR上調(diào)至9 739、3 066、1 263倍;PXR上調(diào)至455、506、298倍;Western blot結(jié)果,地格達(dá)-4各給藥組對PXR具有顯著的下調(diào)、CAR具有顯著的上調(diào)作用(P0.01)。結(jié)論:幽門結(jié)扎能引起急性肝損傷,且能改變對核受體PXR和CAR表達(dá)的變化。地格達(dá)-4通過PXR和CAR膜蛋白表達(dá)變化,引起保護(hù)肝臟,促進(jìn)膽汁分泌功能。
[Abstract]:Objective: to study the effect of Digelda-4 on the treatment of pyloric ligation of liver injury through the change of expression of PXRG and CAR.Methods: 50 rats were divided into 5 groups: normal group and model group. The rat liver injury model was established by pyloric ligation in 3 doses of digerda-4 (1.86 ~ 3.71 ~ 7.43 g 路kg ~ (-1) 路kg ~ (-1)). The changes of PXRCAR expression in rat liver and the sensitivity of liver injury in serum and liver tissue were detected by real-time fluorescence quantitative chain polymerase reaction (RTPCRN) and Western blot assay after intragastric administration for 15 days. Results: compared with the normal group, the expression of PXRCAR and the sensitivity of PXRCAR in serum and liver tissue were measured. In the model group, the serum TBA of alt BILD _ 2 and BILT _ 3 increased significantly, and the GSH-PX of liver tissue decreased significantly (P _ (0.01), P _ (0.01) P _ (0.01)), the peritoneal membrane had obvious yellow color, hydrocele, yellowish urine and naked eye to observe liver congestion, morphological observation showed that hepatic cells had obvious fat change and edema. Compared with the model group, the TBA of BILD _ 2 and BILT _ 3 in each administration group decreased significantly (P 0.01) and GSH-PX increased significantly (P 0.01). The morphology of abdominal cavity and liver was improved by RT-PCR. CAR of nuclear receptor was up-regulated to 9 739 ~ 3 066 ~ 1 263 times. The results of Western blot showed that digerda-4 could significantly down-regulate the expression of PXR. Conclusion: pyloric ligation can induce acute liver injury. Digoda-4 can protect the liver and promote bile secretion through the changes of PXR and CAR membrane protein expression.
